Objective:The aim of the study is to investigate the effects of cigarette smoke extract(CSE)on the polarization of monocytes THP-1 cells and the effects of CSE-induced tumor-associated macrophages(TAMs)on the invasion and metastasis of non-small-cell lung cancer(NSCLC)A549 and PC-9cells.Method:Flow cytometry and qRT-PCR were detected the scavenger receptor level of CD163 and the mRNA levels of IL-10,IL-12,TNF-? and TGF-? in THP-1 cells with different concentration of CSE treatment for 96 h,respectively.Western blot was used to detect the activation of the p-STAT6/STAT6 pathway in THP-1 cells with different concentration of CSE treatment for 96 h.The migration and invasion ability and the expression levels of E-cadherin and Vimentin in A549/PC-9 were detected,following treatments with 4% CSE,or with macrophages co-cultured,or with 4%CSE plus macrophages co-cultured.Transwell assay and Western blot were used to detect the migration and invasion ability and the expression levels of E-cadherin and Vimentin in PC-9 and A549 cells.Results:qRT-PCR showed that the mRNA levels of IL-10,IL-12 and TNF-?decreased,and the mRNA levels of TGF-? increased(P<0.05).Flow cytometry demonstrated that the scavenger receptor CD163 was increased,after treating different concentration of CSE for 96h(P<0.05).Western blot showed that the M2 polarized pathway of p-STAT6/STAT6 was significantly activated by CSE.Compared with blank group,the expression levels of E-cadherin decreased,while the expression levels of Vimentin increased,after co-cultured with CSE pretreated macrophages and 4% CSE plus macrophages co-cultured(P<0.05).Conclusion:1.CSE can induce M2 macrophages in THP-1 cells.2.CSE promotes M2 macrophage polarization by activating p-STAT6/STAT6 signaling.3.CSE-induced M2 polarized macrophages improve the invasion and migration ability of A549 and PC-9 cells.4.CSE-induced M2 polarized macrophages enhance EMT of A549 and PC-9 cells.
