PART 1 SCREENING AND CONSTRUCTION THE MIR-151A-5P-INHIBITOR OF LUNG CANCER CELL LINEObjective:The study aimed to investigate the expression of mir-151a-5p in Lung adenocarcinoma tissue and different lung adenocarcinoma cell lines,screening the highest one and construction the Mir-151a-5p-inhibitor of Lung Cancer Cell LineMethods:RT-q PCR was used to detect the expression of mir-151a-5P in 16 lung adenocarcinoma tissues and 5 different lung adenocarcinoma cells and the highest one was screened out.Then the selected cell line was transfected with mir-151a-5P-inhibitor.Finally,RT-q PCR was used to verify the transfection efficiency.Results:The expression of mir-151a-5p in human lung adenocarcinoma wassignificantly higher than that in normal lung tissue(P<0.01)and also it was higher in A549 cells than other 4 cells(P < 0.01).After transfection the expression of mir-151a-5p was significantly decreased in mir-151a-5P-inhibitor group compared with NC group(P<0.01).Conclusion:The expression of mir-151a-5p was increased in human lung adenocarcinoma tissue and lung cancer cells,and the mir-151a-5p-inhibitor transfection group of A549 cells were successfully constructed.PART 2 MIR-151A-5P REGULATES THE BIOLOGICAL FUNCTION AND CHEMOSENSITIVITY OF LUNG CANCER CELLSObjective:To provide the new basis for the lung cancer treatment and figure out the effects of mir-151a-5p on the proliferation,migration,invasiveness and chemosensitivity of lung cancer by downregulating the expression of mir-151a-5p in A549 cell.Methods:CCK-8 assay was used to detect the proliferation of mir-151a-5P-inhibitor A549 cells.Migration and invasion of transwell assay were used respectively to detect the migration and invasion abilities of mir-151a-5P-inhibitor A549 cells.Western blot was used to detect the expression of EMT-related proteins after inhibitor transfection.CCK-8 and flow cytometry were used to detect the changes of IC50 and apoptotic rate after transfection.Results:Compared with nc group,the proliferative,migratory,invasive and colonal abilities of mir-151a-5p inhibitor group were all decreased(P<0.05).Western blot analysis showed that the expression of E-cadherin was increased in the mir-151a-5p inhibitor transfection group compared withthe NC group,while the expression of N-candherin,Vimentin and Notch1 were decreased(P < 0.01).CCK-8 measurements showed that IC50 in mir-151a-5p inhibitor transfection group was lower than that in NC group(P<0.01).Finally,the results of apoptosis experiment demonstrated that the apoptosis rate of mir-151a-5p inhibitor transfection group was higher than that of NC group(P<0.01).Conclusion:Downregulating the expression of mir-151a-5p in A549 cells can inhibit the proliferation,migration and invasion of lung cancer cells,and increase the chemosensitivity to cisplatin.
