Objective: G protein-coupled receptors(GPCRs)are the largest family of membrane receptors in the human body and are involved in a variety of physiological and pathological functions.GPR183 plays an important role in normal hematopoiesis,but it is unclear whether it is involved in the development of acute T-lymphocytic leukemia(T-ALL).By analyzing the expression of GPR183 in T-ALL and constructing the T-ALL Cutll1 cell line overexpressing GPR183,the effect of GPR183 on the proliferation and apoptosis of Cutll1 cells was devised,and the molecu Les of GPR183 promoting apoptosis of T-ALL Cutll1 cells were initially explored.mechanism.Methods: The GPCR gene differentially expressed in children's T-ALL cells and normal children's T cells was detected by transcriptome sequencing(RNA-seq),and the expression of GPR183 in 13 patients and 4 control groups was verified by q-PCR and two GSE databases.WB verified the expression of GPR183 in two control and four T-ALL cell lines,constructed overexpressing GPR183 lentiviral vector,packaged virus infected Cutll1 cells,and analyzed the expression of GPR183 after virus-infected cells by q-PCR and Western blot;CCK-8,Annexin V,Hoechest,PI(Propidium Iodide)staining was used to detect the effect of GPR183 on the function of Cutll1 cells.Results: The expression of GPR183 in T-ALL was significantly decreased in the RNA-seq data.The expression of GPR183 in the two T-ALL GSE databases of GSE26713 and GSE66638 was consistent with the sequencing results in 13 children's T-ALL samples and 4 T-ALL cells,respectively.The expression of GPR183 m RNA and protein was also confirmed to be significantly decreased in T-ALL.The overexpressed GPR183 lentiviral vector was successfully constructed and the overexpressed GPR183 Cutll1 cell line was established.The growth of Cutll1 cells was significantly decreased after overexpression of GPR183(p< 0.001),apoptosis was significantly increased(p < 0.01),but there was no significant change in cell cycle.Conclusion: The expression of GPR183 was abnormally decreased in children's T-ALL cells.It was initially shown that GPR183 can inhibit the growth of Cutll1 cells and promote cell apoptosis,which may play an important role in the development of T-ALL.
