Part I Expression of serum exosomes ERG,SPDEF and PSMA in patients with prostate cancerObjective:This study is to explore the diagnostic value of the combined detection of exosomal ERG,SPDEF and PSMA in plasma as a promising noninvasive screening tool for prostate cancer.Methods:1.Exosomes derived from the serum of prostate cancer,benign prostatic hyperplasia and health controls.2.Real-time quantitative polymerase chain reaction(RT-PCR)was used to detect the expression levels of serum ERG,SPDEF and PSMA in 63 PCa patients,37 benign prostatic hyperplasia patients and 61 health controls.ROC curve was applied to identify the area under the curve.Fisher linear evaluated the diagnostic value of ERG,SPDEF and PSMA in patients with prostate cancer.3.All data in the experiment was presented as average ±standard error.The data were test by Independent Sample T Test.All the statistical analysis was performed using SPSS22.0and P values less than 0.05 were considered to statistically significant.Results:1.Exosomes from the serum of prostate cancer,benign prostatic hyperplasia and health controls was isolated.2.The expression level of exosomal ERG,SPDEF and PSMA in the serum of patients with prostate cancer was significantly higher than that of healthy controls and benign prostatic hyperplasia(P < 0.05).3.Detection of serum exosomal ERG,SPDEF and PSMA in the area under the ROC curve were 0.891,0.875 and 0.887 for the diagnosis of prostate cancer.The combined detection of serum exosomal ERG,SPDEF and PSMA in the area under the ROC curve was.0.93 for the diagnosis of prostate cancer.4.Fisher linear discriminant function was applied to 161 samples.The correct classification of genes was 86.3%.Conclusion:Exosomes were rapidly and conveniently isolated from the serum of prostate patients by using exo RNasey Serum/Plasma Maxi Kit.Exosomal ERG,SPDEF and PSMA were a potential non-invasive biomarker for PCa.The Combined detection of serum exosomal ERG,SPDEF and PSMA may improved the sensitivity and specifity in diagnosis PCaPart II The relationship between serum exosomal ERG,SPDEF,PSMA and Clinicopathology in patients with prostate cancerObjective The exosomal ERG,SPDEF and PSMA have been reported to be aberrant expression in patients with PCa in the first part,but their relationship with clinicopathological features in PCa is poorly understood.So this part aimed to elucidate the application value of ERG,SPDEF and PSMA for diagnosis and prognosis in PCa.Methods The expression of exosomal ERG,SPDEF and PSMA in 63 patients with prostate cancer were detected by RT-PCR.The clinicopathological data of patients with prostate cancer were collected,including age,PSA level,Gleason score,WHO Grade,TNM stage,lymph node metastasis and so on.The correlation between the expression of exosomal RNA and clinical pathology was analyzed.by SPSS22.0Results The expression levels of serum exosomal ERG was significantly related to the presence of lymph node metastasis,levels of Gleason and levels of T-staging.The expression levels of serum exosomal PSMA was significantly related to the presence of of distant metastasis.There was no significant difference between SPDEF and other indicators.Conclusion It indicated that expression of exosomal ERG and PSMA could reflect the malignancy of PCa in lymph node metastasis,levels of Gleason,levels of T-staging and distant metastasis as plasma biomarkers for diagnosis and prognosis of PCa.
