Objectives XPA binding protein 2(XAB2)plays an important role in the process of transcription-coupled nucleotide excision repair.The genetic variants of XAB2 may affect the capability of transcriptional coupling repair and further involved in the development of cancer.This study aims to explore the relationship between the genetic variation of XAB2 and susceptibility to gastric cancer,in order to serve to population screening for genetic susceptibility to gastric cancer.Methods This study was based on a hospital-based case-control study with 500 patients with gastric cancer and 500 healthy controls.TagSNPs were selected using the Haploview software and the TagSNPs were genotyped by the Sequenom MassArray system.Pearson's chi-square test was used to compare the differences in demographic data between gastric cancer and control groups.Multivariate logistic regression model was used to caculate odds ratio(OR)and its 95% confidence interval(CI)to assess the impact of XAB2 variants on the risk of gastric cancer.Non-parametric multifactor dimensionality reduction(MDR)is used to evaluate gene-gene interaction.Results In this study,total 5 TagSNPs(rs4134816,rs4134819,rs4134860,rs794078,and rs794083)were selected to evaluate the association of XAB2 genetic variants with the risk of gastric cancer.We found that the individuals carrying XAB2 rs794078 AA genotype had a lower risk of gastric cancer than those with GG genotype(OR=0.33;95%CI=0.12-0.91,P=0.032).The heterozygous AG genotype was not associated with Gastric cancer risk(OR=1.29;95%CI=0.95-1.76,P=0.105).The other TagSNPs had no any effect on the risk of gastric cancer.Non-parametric multi-factor dimensionality reduction(MDR)analysis shows that XAB2 rs794078 is the best interaction model with test balance accuracy of 51.68%,cross-check consistency of 9(P=0.055).When conducted stratified analysis,we found that smokers with XAB2 rs794078 AA genotype had a significantly lower risk of gastric cancer compared with non-smokers with GG genotype(OR=0.11;95%CI=0.01-0.91;P=0.040.Conclusions The XAB2 tagSNP rs794078 AA genotype is a protective factor for gastric cancer.This study demonstrates that XAB2 plays an important role in the development of gastric cancer.Figure5;Table13;Reference 141...
