Objectives To observe the effect of self-made Qingre Liangxue recipe on the expression of superoxide dismutase(SOD)and malondialdehyde(MDA)in serum of rats with acute radiation enteritis,interleukin-6(IL-6),tumor necrosis factor(TNF-a)and interleukin-10(IL-10)in intestinal tissue of rats with acute radiation enteritis.Furthermore,it provides a more reliable experimental basis for the clinical application of Qingre Liangxue recipe in the prevention and treatment of acute radiation enteritis.Methods 60 male SD rats were selected and fed adaptively for 3 days and fasting for 12 hours.after weighing,they were randomly divided into normal group(n = 8)and model group(n = 52).Except the normal group,all rats were fixed with 10% chloral hydrate2.5ml/Kg after general anesthesia.Simens PRIMUS linear accelerator was used to irradiate the above rats with 6MV high energy X-ray.The irradiation range was 1.5cm×1.5 cm on the pubic symphysis,and the lead block obscured the non-irradiated area.Single irradiation,source skin distance 100 cm,dose rate 200 c Gy/min,total dose 12 Gy,fasting water 8hours before irradiation.body weight,mental state,frequency of defecation and nature of defecation were recorded after irradiation.The criteria for the success of the model were as follows: the irradiated rats appeared yellowing or stool changes within 1 day,such as mucus stool,hematochezia and other radiation injury symptoms.after the occurrence of related symptoms,2 of the 52 rats in the model group were randomly selected.The rats were killed for cervical vertebra removal.a small intestine about 5cm length was dissected and the histopathological changes of the small intestine were observed under light microscope to confirm the success of the model.The rats were randomly divided into model group,western medicine control group,traditional Chinese medicine perfusion group,traditional Chinese medicine enema group,traditional Chinese medicine gavage +enema group.According to the conversion method of human and animal body surface area,the original drug dose of each rat was determined by gavage or enema,and the decoction was prepared by self-made Qingre Liangxue decoction,which was given to three traditional Chinese medicine groups according to the dose of 2g/Kg body weight.The western medicine control group was given the corresponding dose of dexamethasone,and the normal group and the model group were given the same amount of normal saline.The rats were treated once every other day for 4 weeks.the body weight,general condition and death of rats were recorded during the treatment.Four weeks later,the rats were killed and the levels of superoxide dismutase(SOD)and malondialdehyde(MDA)in blood and interleukin-6(IL-6),interleukin-10(IL-10)and tumor necrosis factor-a(TNF-a)in rectal tissue were measured.At the same time,the tissue specimens were sent to confirm the histological changes.Results 1 Finally,55 rats were included as the experimental subjects,including 8 rats in the normal group,10 rats in the model group,9 rats in the western medicine control group,9 rats in the traditional Chinese medicine enema group,10 rats in the traditional Chinese medicine enema group and 9 rats in the traditional Chinese medicine gavage + enema group.2 General situation of rats: compared with the normal group,the rats in each group had increased body weight,decreased activity,decreased diet,dim hair color,and increased times of blood stool and mucous stool.3 There was no significant difference in body weight among the three groups before the establishment of the model(P <0.05).After treatment,the weight of the rats in the model group,the western medicine control group and the traditional Chinese medicine group was significantly lower than that in the normal group(P < 0.05),and the quality of the rats in the three groups was higher than that in the western medicine control group(P < 0.05).There was no significant difference in body mass among the three groups(P > 0.05).4 Compared with the normal group,the activity of SOD in serum decreased(P < 0.05),MDA content increased(P <0.05),the contents of TNF-a and IL-6 in intestinal tissue increased(P < 0.05),and the content of Pl in intestinal tissue increased(P < 0.05)in the model group compared with the normal group.The content of IL-10 decreased(P < 0.05).5 Compared with the model group,the activity of SOD in the serum of the three groups of traditional Chinese medicine and the control group was increased(P < 0.05),MDA content decreased(P < 0.05);The contents of TNF-a and IL-6 in intestinal tissue decreased(P < 0.05),while the content of IL-10 increased(P<0.05).6 Compared with the western medicine control group,the activity of SOD in serum of the three groups of traditional Chinese medicine was higher than that of the western medicine control group,and the content of MDA was lower than that of the western medicine control group.The contents of TNF-a and IL-6 in intestinal tissue were lower than those in the western medicine control group,and the contents of IL-10 in the intestinal tissue were lower than those in the western medicine control group.7There was no significant difference in the levels of SOD,MDA in serum and TNF-a,IL-6and IL-10 in intestinal tissue among the three groups(P > 0.05).Conclusions 1 Qingre Liangxue recipe can increase the activity of SOD in serum,decrease the content of MDA,eliminate oxygen free radicals,inhibit lipid peroxidation,reduce the peroxidation of intestinal mucosa and promote the recovery of antioxidant system in rats with radiation enteritis.Speeding up the repair of intestinal tissue injury.2Qingre Liangxue recipe can up-regulate the level of IL-10 in intestinal tissue,decrease the content of TNF-a and IL-6,reduce the inflammatory reaction in intestinal tract,reduce intestinal injury and promote the repair of damaged intestinal mucosa
