Objective: To explore the survival rate of different layers of lipoaspirate fractions centrifuged by Coleman technique,in order to guide clinical application better.Methods:The inguinal fat tissue of New Zealand rabbits was harvested and centrifuged by Coleman procedure.Then,the centrifuged lipoaspirate was divided into upper,middle,and lower layers and transplanted into the back of rabbits.Three months later,the survival volume of the fat grafts were measured.The number of vesicles,the degree of fibrosis and the integrity of the fat cells of the fat grafts were observed by HE staining.The neovascular density of the fat grafts were observed by immunohistochemical staining for CD31.Result:1)The survival volume of the lower layers fat grafts were significantly higher than that of the upper and middle layers fat grafts,and the differences were statistically significant(P<0.05).There was not significant difference between the upper layers and middle layers fat grafts(P>0.05).2)The number of vesicles and fibrosis of lower fat grafts were lower than those of upper and middle layers fat grafts andthe number of intact fat cells was higher than that of other layers,and the differences were statistically significant(P<0.05).There were not significant differences in the number of vesicles,degree of fibrosis and the number of intact fat cells between upper and middle layers fat grafts(P>0.05).3)The vascular density of the lower layers fat grafts were significantly higher than that of the upper and middle layers fat grafts,and the difference was statistically significant(P<0.05).There was not significant difference in the vascular density between the upper and middle fat grafts.(P>0.05).Conclusion:The lower layer lipoaspirate fractions centrifuged by Coleman technique could be an ideal candidate for fat grafts due to it better survival rate and quality after centrifuged...
