Objectives: Establishing SE models on mice,and treating them with high or low dose of borneol.Try to reveal the molecular mechanism of aromatic resuscitation therapy through detecting the expression of TRPV1 protein,TRPA1 protein and GABA transmitter in mice’s hippocampus.Methods: 96 male C57BL/6J mice were divided into 4 groups randomly.There are 6 mice in control group,30 in SE model group,30 in low-dose borneol group,30 in high-dose borneol group.SE was induced by pilocarpine hydrochloride.To observe the animal behavior,stain the hippocampus of mice with Nissl staining,detect the expression amounts of TRPV1 protein,TRPA1 protein and GABA transmitter in different parts of hippocampus by immunohistochemistry.Results:1.Animal behavior: The SE grades and degrees of mice treated with natural borneol decreased(P<0.05,P<0.01).2.Cell morphology: The damages of hippocampal neuron are showed from low to high degrees as follows: control group,high-dose borneol group,low-dose borneol group,SE model group.3.Immunohistochemistry3.1 Comparisons of the average optical density of TRPV1 protein in the hippocampal CA1 and CA3 regions of each groupCA1: Compared with control group,the density of TRPV1 protein of SE model group and low-dose borneol group increased significantly(P<0.01),high-dose borneol group shows no significant difference(P>0.05).Compared with SE model group,the density of TRPV1 protein of low-dose borneol group and high-dose borneol group decreased significantly(P<0.01).Compared with low-dose borneol group,high-dose borneol group decreased significantly(P<0.01).The results of CA3 are the same as CA1.3.2 Comparisons of the average optical density of TRPA1 protein in hippocampal CA1 and CA3 regions of each groupCA1: Compared with control group,the density of TRPA1 protein in SE model group,low-dose and high-dose borneol group increased significantly(P<0.01,P<0.05).Compared with SE model group,low-dose borneol group and high-dose borneol group decreased significantly(P<0.01,P<0.05).Compared with low-dose borneol group,high-dose borneol group shows no difference(P>0.05).The results of CA3 are the same as CA1.3.3 Comparisons of the average optical density of GABA in the hippocampal CA1 and CA3 regions of each groupCompared with control group,the average optical density of GABA in SE group decreased significantly(P<0.01),which in high-dose borneol group increased significantly(P<0.01).Compared with SE group,low-dose borneol group and high-dose borneol group increased significantly(P<0.01).Compared with low-dose borneol group,high-dose borneol group increased significantly(P<0.01).The results of the CA3 are same as those of the CA1.Conclusions:1.It shows inhibition of natural Borneol to SE mice,according to prolonging latency of SE,decreasing seizure level and damage of hippocampal neurons.2.It is related to the regulation of TRPA1 and TRPV1 protein expressions in hippocampal CA1 and CA3 regions and the release of neurotransmitter GABA that borneol inhibiting SE.It is one of the molecular mechanisms that borneol plays an aromatic resuscitation role.
