Objective To investigate the effects of Feixinning Decoction on the expression of matrix metalloproteinase-9(MMP-9)and matrix metalloproteinase inhibitor-1(TIMP-1)in a rat model of chronic obstructive pulmonary disease(COPD).The therapeutic effect and mechanism of Feixinning Decoction on COPD rats.Methods Sixty Wistar rats were randomly divided into 6 groups: normal group,COPD model group,prednisone acetate group,and low-,medium-,and high-dose groups of Feixinning Decoction,with 10 rats in each group.Except the normal group,the other 5 groups replicated the COPD model by instilling lipopolysaccharide(LPS)into the airway,and the modeling period was 90 days.After the modeling,the normal group,the COPD model group was given an equal volume of distilled water,the prednisone acetate group was given 0.027g/Kg of prednisone acetate tablets,and the low,medium and high dose groups were given different doses of Feixinning Decoction(3.3ml/kg、6.6ml/kg、13.2ml/kg),for 28 days.At the end of the experiment,the rats were sacrificed and blood was taken by abdominal aorta.The expression of MMP-9 and TIMP-1 in the serum of rats was detected by ELISA.HE staining was performed in the right lower lobe of rats to observe the morphology of lung tissue.Structural and pathological changes were performed by Real-Time PCR to detect mRNA expression in lung tissue of rats.Results 1.HE staining results showed that Feixinning could improve lung tissue damage in COPD rats to some extent.2.The results of ELISA showed that the expression levels of MMP-9 and TIMP-1 in the other 5 groups were significantly higher than those in the normal group(P<0.05).Compared with the COPD model group,MMP-9 and TIMP in each treatment group.The expression level of-1 wasdecreased(P<0.05): the prednisone acetate group was the most significant(P<0.05),and the high dose group of Feixinning was second only to the prednisone acetate group(P<0.05).The results of Real-Time PCR showed that the expression levels of MMP-9 and TIMP-1 in the other five groups were significantly higher than those in the normal group(P<0.05).Compared with the COPD model group,in each treatment group,MMP-9,TIMP-1 expression level decreased(P<0.05): the prednisone acetate group decreased significantly(P<0.05),and the Feixinning high dose group(P<0.05).Conclusion Feixinning Decoction can reduce airway inflammation and interfere with airway remodeling by inhibiting the expression of MMP-9 and TIMP-1 in COPD rats.The improvement of airway remodeling in COPD may be related to dose.
