In this study,the phenolic compounds and its related antioxidant capacities of Chuanminshen violaceum roots and leaves collected from 4 major producers including Huaikou,Wuma,Yuxi and Caangxi were investigated,as well as their primary structure and antioxidant capacities,hypoglycemic effects and immunomodulating activities.Meanwhile,the differences of functional compoents and its related bioactivities between the Chuanminshen violaceum from different locations were analyzed.And the results are as below:1.Based on the ultrasound assisted extraction(UAE)method,the effect of methanol concentration,ratio of raw materials to liquid,UAE time and UAE temperature on the total phenolic content of Chuanminshen violaceum root was investigated,and the method was subsequently optimized.Results showed that the significance of above factors was indicated as ratio of raw materials to liquid > methanol concentration > UAE time > UAE temperature.Meanwhile,under the optimized conditions as methanol concentration of 80%,ratio of raw materials to liquid of 40 mL/g,UAE time of 100 min and UAE temperature of 50℃,the total phenolic content of sample was 5.999±0.19 mg GAE/g.2.The difference of phenolic compunds content and its related antioxidant capacities of Chuanminshen violaceum roots and leaves from different locations was investigated and compared.Results showed the significant difference of total phenolic content(TPC),total flavonoid content,chlorogenic acid content and rutin content between the Chuanminshen violaceum roots and leaves from different locations,which is Yuxi > Wuma > Huaikou > Cangxi.Meanwhile,these above values of Chuanminshen violaceum leaves are significantly higher than thoese of Chuanminshen violaceum roots,and the same tend of difference of antioxidant capacities between Chuanminshen violaceum roots and leaves from different locations was observed.Furthermore,according to the results of Pearson’s analysis,the phenolic compunds in Chuanminshen violaceum were indicated high corerelations of antioxidant capacities.3.The difference of primary structure and related bioactivities of crude polysaccharides of Chuanminshen violaceum roots and leaves from different locations was investigated and compared.Results showed the existence of acid polysaccharides in both Chuanminshen violaceum roots and leaves,and the primary structures of crude polysaccharides of different part of Chuanminshen violaceum are different.Chuanminshen violaceum roots polysaccharides have less uronic acid contents,and the major monosaccharide composition is GalA,Glc,Gal and Ara,while the molecular weights is higher;leaves polysaccharides have more urnoic acid contents,and the major monosaccharide composition is Man,Rha,GalA,Glc,Gal and Ara,while the molecular weights is lower.The primary structures of crude polysaccharides of Chuanminshen violaceum from different locations are similar.The antioxidant capacities were observed in both crude polysaccharides of Chuanminshen violaceum roots and leaves,however the Chuanminshen violaceum leaves crude polysaccharides showed higher level of antioxidant capacities.Additionally,the crude polysaccharides of Chuanminshen violaceum leaves have high level of hypoglycemic activities,while the crude polysaccharides of Chuanminshen violaceum roots have high level of immunomodulating activities with the tend of Huaikou > Wuma > Jintang > Cangxi.
