Objective:To explore antibacterial activity and cytotoxicity of AgHA and Bio-Oss in order to find the suitable quantity of Ag in AgHA and the best mixture ratio of bone substitutive material which contained AgHA and Bio-Oss,which can provide another resolution of peri implantitisMethods:AgHA with different amount of silver was synthesized by chemical precipitation method in this study.Then,Porphyromonas gingivalis(Pg)and Fusobacterium nucleatum(Fn)which are the main bacteria in plaque during peri-implantitis were used to test the antibacterial activity of AgHA.And the suitable quantity of Ag could be chosen according to antibacterial activity and cytotoxicity Then the suitable AgHA was mixed Bio-Oss at ratios of 1:1,1:2 and 1:4 by weight Minimum bactericidal concentration(MBC)and short-term antimicrobial efficacy were measured.And we observed their effects on the expression of bacterial virulence genes(FimA for Pg and FadA for Fn),and human periodontal ligament fibroblasts and rat bone marrow stromal cells were used to measure the cytotoxicity.This study was to explore the suitable quantity of Ag in AgHA and the best mixture ratio of bone substitutive material which contained AgHA and Bio-Oss through the tests of antibacterial activity and cytotoxicityResults:The inhibitory effect of three kinds of AgHA on bacteria is proportional to silver content,and cytotoxicity of 0.7AgHA was significantly higher than that of 0.5AgHA and 0.3AgHA.While there was no significant difference between the 0.5AgHA and 0.3AgHA.When talking about the mixture,antibacterial activities of group 1:1 is roughly similar to that of group 1:2,which were higher than that of group 1:4;cytotoxicity of group 1:2 was similar to that of group 1:4,which was lower than that of group 1:1Conclusions:According to antibacterial performance and cytotoxicity,0.5AgHA was the most suitable silver content.When talking about the mixture,mixing 0.5 AgHA with Bio-Oss(?)at a ratio of 1:2 by weight was recommend.
