ObjectiveStudy the effect of extremely low frequency electromagnetic fields(ELF-EMF)on the matrix metalloproteinase-2(MMP-2)activity,collagen type I(Collagen I)synthesis in scleral fibroblasts(SF)cells and the proliferation rate,expression and secretion of transforming growth factor-?2(TGF-?2)in retinal pigment epithelium(RPE)cells.Study the intervention effect of puerarin in these processes.MethodsMMP-2 activity were measured by gelatin enzymography.MMP-2 and Collagen I protein expression were detected by Western-Blot.Proliferation rate of RPE cells were quantitated by cell counting kit-8 assay.TGF-?2 mRNA expression and secretion were measured by Real-Time PCR and ELISA analysis,respectively.ResultsCompared with control group,MMP-2 activity and protein expression increased and Collagen I protein expression decreased in radiation group.Compared with radiation group,MMP-2 activity and protein expression decreased and Collagen I protein expression increased in inhibitor group.In puerarin group,MMP-2 activity and protein expression decreased and Collagen I protein expression increased compared with radiation group.In radiation group,viability of RPE cells were inhibited and TGF-?2 mRNA expression and secretion in RPE cells increased compared with control group.In puerarin group,viability of RPE cells enhanced and TGF-?2 mRNA expression and secretion in RPE cells reduced compared with radiation group.The difference was statistically significant.ConclusionELF-EMF may reduce the expression of Collagen I by stimulating the activity and expression of MMP-2 in SF cells.MMP-2 inhibitor and Puerarin can inhibit the activity and expression of MMP-2 in SF cells and reduce the degradation of Collagen I.ELF-EMF inhibited the viability of RPE cells and increased the expression and secretion of TGF-?2 in RPE cells.Puerarin reversed the action to some extent.
