Traditional pharmacology research is limited to ‘single drug to single target’.Although plenty of relationship between drugs and diseases has been understood deeply,little progress has been made in studies on some kinds of traditional Chinese medicine(TCM).Most TCM contains a huge source of chemical constituents which can interact with a great deal of targets.Thus,it is difficult to study TCM in depth with traditional way in pharmacology research.Network pharmacology observes the effects of drugs on disease networks systematically based on the network of diseases,genes,targets and drugs,which coincides with the theory of Whole View in Chinese medicine.It can be said that the emergence of network pharmacology provides a new solution for the modernization of TCM.Besides network pharmacology,the have been some other new methods applied in the research TCM in these years.And these new methods inject new life into the research of TCM.Leonurus japonicus(Yimucao),Coicis semen(Yiyiren),Sophora tonkinensis(Shandougen)and Curcumae rhizoma(Ezhu)have the effect of prevention and treatment of oral cancer at different levels according to literatures.The study constructed Yimucao,Yiyiren,Shandougen and Ezhu’s component-target networks and oral cancer-related targets network based on network pharmacology.Then the TCM component-target networks were merged with the oral cancer-related targets network to build four TCM’s component-target-oral cancer-related targets networks.Cluster analysis was applied to gain subnetworks of the TCM’s component-target-oral cancer-related targets networks.Targets involved in the subnetworks were recorded as key targets.Then we performed GO and KEGG pathway enrichment analysis to clarify the mechanism of the four kinds of TCM preventing and treating oral cancer.At last the electronic tongue was taken for an example to introduce other new methods applied in the research of TCM.The main results are as follows:1.We retrieved Chinese herbal components from TCMSP database,including 51,38,79 and 81 from the four herbs: Yimucao,Yiyiren,Shandougen and Ezhu.2.We explored the effective targets of the four kinds of TCM from the database of TCMSP,Swiss Taget Prediction and STITCH.The number of these targets in Yimucao,Yiyiren,Shandougen,and Ezhu was 415,273,463,and 256 respectively.3.Component-target netwoks of Yimucao,Yiyiren,Shandougen and Ezhu were constructed with Cytoscape.4.19 oral cancer-related targets were collected and used to contruct oral cancer-related network with Bisogenet,a Cytoscape plugin.5.Four component-target networks were merged with the oral cancer-related network respectively to build TCM component-target-oral cancer-related target netwotks.6.MCODE,another Cytoscape plugin,was used to make cluster analysis of the component-target-oral cancer-related target networks.Then the targets in the subnetworks were recorded as key targets.7.We performed GO and KEGG pathway enrichment analysis for the key targets of the herbs.The number of function four herbs’ key targets involving was 73(Yimucao),36(Yiyiren),37(Shandougen)and 43(Ezhu);the number of pathways four herbs’ key targets involving was 26(Yimucao),7(Yiyiren),27(Shandougen)and 26(Ezhu).8.The application in the TCM research of the electronic tongue was introduced.And we used an electronic tongue to discriminate and predict samples of Cynomorium songaricum Rupr.from different growing areas in China.The study used network pharmacology to explore the mechanism of the four kinds of TCM preventing and treating oral cancer successfully.And the electronic tongue was taken for an example to introduce other new methods applied in the research of TCM.It is helpful for researchers to strengthen the understanding of network pharmacology and oral cancer.Besides,it provides new ideas for studying TCM.
