Objective: To isolate and purify the chemical constituents from the 30% ethanol fraction of Tibetan medicine Auricularia auricula,identify monomer compounds,and study the XOD inhibitory activity and antioxidant activity of the compounds in vitro.Methods:(1)Separation and purification of the chemical components of 30% ethanol by silica gel column chromatography,Sephadex LH-20,HPLC,etc.,according to their physicochemical properties and modern spectroscopy techniques(including hydrogen spectrum carbon spectrum,UV,IR,mass spectrometry,etc.)were used to identify the structure of these compounds.(2)Using XOD as the target and using astragalus as the substrate,the optimal reaction conditions for the binding of each compound to XOD were screened in vitro,and the reliability of the reaction conditions was verified by the positive drug allopurinol.The obtained compound was subjected to an experiment for XOD inhibitory activity under the above reaction conditions.(3)The antioxidant activity of the isolated and purified compounds in Brachypodium serrata was evaluated by DPPH method and ABTS method.Results:(1)A total of 17 compounds were identified from the 30% ethanol fraction of the ethyl acetate extract of Brachypodium serrata,6 phenylethanoid glycosides: lagotiside B(2),hemiphroside B(3),plantainoside D(4),Plantamajoside(5),2'-O-acetylplantamajoside(6),acteoside(7);5 iridoid glycosides: Scutellariosid II(8),Globularimin(9)Baldaccioside(10),10-O-trans-isoferheyl sterol(11),8-epi-loganic acid(12);3 flavonoids: luteolin(13),luteolin-7-O-?-D-glucopyranoside(14),wheat flavin 7-O-?-D-glucopyranoside(15),three other compounds: lagotisideC(1),benzyl alcohol-O-(2 '-O-?-D-xylopyranosyl)-?-D-glucopyranoside(16),cinnamic acid(17).Among them,1,2 are new compounds;3,4,6,10,11,16 are isolated for the first time;5,7,8,9,12 are the first separation of Brassica chinensis.(2)Compounds 4,13,and 14 have a good inhibitory effect on XOD activity,and compound 13 has no significant difference in the ability to inhibit XOD activity and positive drug;compound 14 has a glucose in addition to compound 13,although there is significant inhibitory activity.The ability is weaker than that of Compound 13;Compound 4 also has a strong ability to inhibit XOD activity.Compounds 1,2,3,5,6,7,10,12,16,17 have weak ability to inhibit XOD activity with the above three compounds.(3)Compounds 3,4,11,and 13 have strong ability to scavenge DPPH,and 1,2,5,6,7,8,9,10,12,15,16,and 17 have weaker ability to scavenge DPPH.Compounds 3,4,11,13,and 14 have strong ability to clear ABTS,and compounds 9,10 have a slightly stronger ability to clear ABTS.Compounds 1,2,5,6,7,8,12,15,16 The ability to clear ABTS is weak.Conclusion: This study studied the chemical constituents of 30% ethanol in A.sinensis,and studied its XOD inhibitory activity and the antioxidant activity of the compound.The chemical constituents of 30% ethanol of Brachypodium serrata were isolated,and 17 compounds were obtained.Three of them had strong inhibitory effect on XOD.The IC50 value of antioxidant activity of the four compounds was lower than that of the positive drug VC,which showed that it was highly oxidizing.For the positive drug VC.This study provides a basis for the study of anti-gout drugs,and the selected compounds can be used as natural antioxidants to reduce the side effects of synthetic oxidants and provide a theoretical basis for clinical applications.
