Objective To analyze the expression of long non-coding RNA BC200 in NSCLC and the contact between BC200 and clinicopathological features,and to explore its clinical significance and its relationship with epithelial-mesenchymal transformation.Methods1.Samples: We collected lung cancer tissue and serum specimens of NSCLC patients who underwent surgery in Tianjin Chest Hospital from 2014 to 2018.2.The screening stage: 1,158 annotated lncRNAs and 2,869 non-annotated lncRNAs were selected from 35 human lung cancer cell lines in the preliminary experiment,and 13 non-annotated lnc RNAs with abnormal expression were screened through COPA(cancer outlier profile analysis)technology,and 4 new lnc RNAs were selected as candidates.Then,QPCR was used to analyze the expressions of lnc RNAs in 35 cases of NSCLC and 22 cases of adjacent normal tissues.3.The validation phase: we selected lncRNA which had statistically significance and the best diagnostic performance.We used q RT-PCR to detect 60 cases of NSCLC and 60 cases of adjacent normal tissues as well as 40 cases of NSCLC and 40 cases of adjacent normal tissues of EMT related protein expression.Then detect the expression of lnc RNA in serum samples.4.Statistial analyses: We use SPSS 22.0 statistical software to analyze the data.T-test or Mann-Whitney U test were used to compare the data among the measurement groups,chi-square test was used to compare the counting data,pearson correlation coefficient was used for the correlation analysis,and the diagnostic efficacy was described by the ROC curve.Results1.In the screening stage,LCAL1 and BC200 were selected.2.Expression of BC200 and EMT-related proteins in lung cancer.The expression level of BC200,N-cadherin and Snail m RNA in the lung cancer tissues were higher than that in the normal samples,and the expression level of E-cadherin m RNA in the cancer tissues was significantly lower than that in the adjacent tissues (P<0.05).The expression level of BC200 in non-small cell lung cancer serum was higher than that in healthy subjects.3.Immunohistochemical results of EMT-related proteins.E-cadherin was lack in cancer tissue,N-cadherin and Snail were high expression in the cancer tissue,E-cadherin,N-cadherin and Snail positive expression rate in NSCLC tissues were40%(16/40),57.5%(23/40),57.5%(23/40),the positive expression rate of normal lung tissues were 95%(19/20),5%(1/20),10%(2/20).4.The expression of BC200 and Snail m RNA in NSCLC tissues was negatively correlated with E-cadherin m RNA(r=-0.31,r=-0.313)and the BC200 positively correlated with Snail and N-cadherin m RNA(r=0.305,r=0.257).The high expression of BC200 in lung cancer was correlated with lymph node metastasis,clinical stage,E-cadherin,N-cadherin and Snail positive rate.5.Diagnostic value of BC200 for lung cancer.The area under the curve is 0.747.The area under the curve in serum was 0.659.Conclusions1.BC200,N-cadherin and Snail m RNA expression in lung cancer tissues were higher than that in the adjacent normal tissues,and the expression level of E-cadherin m RNA in the cancer tissues was lower than that in the adjacent tissues.It was suggested that lnc RNA BC200 and EMT related proteins were related to the occurrence of NSCLC.2.Those with negative E-cadherin,N-cadherin and Snail positive showed high BC200 expression,suggesting that lnc RNA BC200 was related to EMT-related proteins.3.The expression level of BC200 in lung cancer tissues was correlated with lymph node metastasis,clinical stage,It was suggested that BC200 may be associated with cancer cell invasion and metastasis in NSCLC.4.LncRNA BC200 was upregulated in tissues and serum of patients with NSCLC,which had a good value in the diagnosis of NSCLC and was a potential diagnostic marker for NSCLC.
