Nanomedicine has been widely used in the research of tumor therapy.The nanomedicine carriers loaded with traditional chemotherapeutic drugs can improve the tumor suppressing effect and reduce the toxic side effects.Polymer carriers are commonly used drug carriers in the study.Because of their superior biocompatibility,flexible drug loading methods,suitable size,and functional modification,a variety of related nanomedicine have entered the clinical test.As a novel treatment method,photothermal therapy has the advantages of low toxic and side effects,low invasiveness,and remarkable therapeutic effect,and is increasingly applied to the research of tumor treatment.The polymer carriers with photothermal conversion ability can achieve the synergistic effect of chemotherapy and photothermotherapy.Firsrt,we prepared dual drug-targeted nanoparticles,loaded with chemotherapeutic drugs docetaxel(DTX)and bortezomib(BTZ).By connecting pH-responsive covalent bonds,we avoided the non-specific binding of bortezomib to various proteins and achieved the pH-responsive drug release.We successfully applied bortezomib,previously used only for lymphoma,to the treatment of solid tumorsBased on the above studies,we have prepared multi-functional nanoparticles for the combined treatment of photothermal therapy and chemotherapy for breast cancer.The nano drug delivery system is loaded with docetaxel(DTX)and doxorubicin(DOX),and is connected with aptamer for targeted recognition of breast cancer.The modified polydopamine has good photothermal conversion capability.Under the irradiation of near-infrared light with a wavelength of 808 nm,the nanoparticles can generate localized high temperatures in tumor tissues and specifically kill tumor cells.Blocking cell autophagy by interfering with lysosomes can reduce the degradation of drug-loaded nanoparticles in tumor cells.In summary,a dual drug-targeted nano-drug delivery system was prepared for multimodal treatment of breast cancer and achieved a better therapeutic effect.
