Objective: To investigate the protective effect and mechanism of Buyang Huanwu Decoction on cerebral vasospasm after subarachnoid hemorrhage.Methods: A model of subarachnoid hemorrhage was established by double injection of occipital pool.Garcia neurological function score was used to score neurological function in rats.HE staining was performed on rat basilar artery tissue,and the diameter of basilar artery was measured.Western blot The expression of PI3 K,Akt,p-PI3 K,and p-Akt protein in basilar artery was detected.The expression of eNOS in basilar artery was detected by immunohistochemistry.The expression of PI3 K,Akt and eNOS mRNA was detected by PT-PCR.The expression of NO ? ET-1was detected by Elisa in rat cerebrospinal fluid.Result:(1)After the second occipital pool was injected,the neurological function score of the rats was significantly decreased,and the neurological function score was significantly increased 7 days after the administration of Buyang Huanwu Decoction;(2)HE staining results showed that Buyang Huanwu Decoction can effectively alleviate cerebral vasospasm after subarachnoid hemorrhage in rats,and the diameter of basilar artery is obviously widened;(3)Western blot analysis showed that the expression of PI3 K,Akt,p-PI3 K and p-Akt protein in the basilar artery of SAH rats was significantly decreased,while the expression of PI3 K,Akt,p-PI3 K and p-Akt protein was significantly increased after treatment with Buyang Huanwu Decoction.(4)The expression of eNOS in the basilar artery of SAH rats was significantly decreased by immunohistochemistry.The expression of eNOS protein was significantly increased after treatment with Buyang Huanwu Decoction.(5)The expression of PI3 K,Akt and eNOS mRNA in the basilar artery of SAH rats was significantly decreased by PT-PCR,while the expression of PI3 K,Akt and eNOS mRNA was significantly increased after treatment with Buyang Huanwu Decoction.(6)The expression of NO in cerebrospinal fluid of SAH rats was significantly decreased by Elisa method.The expression of ET-1 was significantly increased,while the expression of NO in cerebrospinal fluid of rats was significantly increased after treatment with Buyang Huanwu Decoction,and the expression of ET-1 was significantly decreased.Conclusion: Buyang Huanwu Decoction can protect the cerebral vasospasm after subarachnoid hemorrhage through the PI3K/Akt/eNOS signaling pathway and increase NO production.
