Procyanidine(PC),the natural flavonoids,has been reported to possess a variety of biological activities,including anti-oxidant,anti-tumor,and anti-inflammatory.Lead(Pb)is one of the most widespread and insidious environmental toxins,which can induce injury in different organs of the body.In the present study,we have used an experimental model of mice exposed to lead(Pb)as a model of hyperlipidemia and hepatic lipid metabolism disorders.We aimed to determine whether procyanidine can protect mouse from Pb-induced hyperlipidemia.We aimed to reveal the intervention effect of procyanidine on lead-induced hyperlipidemia.The main results are as follows:(1)The effect of grape seed proanthocyanidins on lead-induced hyperlipidemiaThe levels of blood lipid(serum cholesterol,triglycerides,LDL and HLD)were analyzed.The changes of body weight in mice were also observed.The results showed that lead could induce the hyperlipidemia in mice.However,grape seed proanthocyanidins treatment could inhibit the lead-induced hyperlipidemia in mice.(2)Grape seed proanthocyanidins inhibited Pb-induced liver injury and decreased Pb concentrations in miceThe lead levels of blood and liver were detected.The diagnostic indicators of liver damage and the histopathological changes were analyzed.Our results showed that procyanidine significantly Pb concentrations in mice and prevented lead-induced damage in livers of mice.(3)The effects of grape seed proanthocyanidins on lead-induced liver oxidative stress in miceThe levels of MDA,GSH and the activities of antioxidant enzymes(SOD,CAT,GPx,GST,GR)were detected.The results showed that grape seed proanthocyanidins could inhibit Pb-induced oxidative stress and increase could the activities of antioxidant enzymes in livers of mice.(4)The effects of grape seed proanthocyanidins on lead-induced lipid metabolism related gene expressionThe expression levels of LXR?,SREBP-2,HMGR ? CYR7A1 were measured by RT-PCR amd western blot.The results showed that grape seed proanthocyanidins could inhibit the expression levels of LXR?,SREBP-2,HMGR and increase the CYR7A1 expression level in the livers of mice.
