Aim:A rat model of diabetic cardiomyopathy was established.The pathological changes of myocardial tissue and the related indexes of oxidative stress,SOD,MDA,NOS,GSH-PX,NOX4 and p47 PHOX protein factors were observed by using Guizhi Jiagui granule as intervention.Influence and evaluation of the effect of Guizhi Jiagui Granule on diabetic cardiomyopathy.Methods :SD male rats weighing 200±20g were randomly divided into model group,metformin group,Guizhi plus granule high dose group,Guizhijiagui granule middle dose group,Guizhi plus granule low dose group,12 groups in each group.The intraperitoneal injection of streptozotocin solution combined with high-fat diet model was given.The corresponding drugs were given 24 hours after model establishment: blank group and model group,normal saline was administered;metformin group,metformin aqueous solution was administered according to standard,cassia twig In the medicinal dosage group,different amounts of aqueous granules were administered according to the conversion criteria of rats and humans.In the experiment,blood was taken from the tail vein of each group,blood was collected from the abdominal aorta,arterial blood and heart tissue were collected.Blood glucose was measured 2 hours after the meal,biochemical analyzer was used to detect serum biochemical indicators,and HE staining was used to evaluate the pathological changes of heart tissue.Oxidative damage markers and antioxidant levels,WB assay for relative expression of protein factors.Results:1.General changes:(1)Body weight of each group after administration: Compared with the model group,the body weight of the Guizhi plus Guizhong group was significantly reduced,the difference was statistically significant(P<0.05),and the metformin group Compared with the dosage group of GuizhiJiagui,there was no statistical difference(P>0.05).(2)2h postprandial blood glucose: Compared with the model group,the metformin group,Guizhi plus Guizhong high-dose group had significantly reduced blood glucose,and the difference was statistically significant.The significance of learning(P<0.05),compared with the metformin group,there was no statistical difference between the Guizhi plus Gui dosage group(P>0.05).\2.Serological changes:(1)total cholesterol,triglyceride,glycosylated hemoglobin: compared with the model group,metformin group,Guizhi plus Gui high dose group were significantly reduced,the difference was statistically significant(P<0.05)Compared with the metformin group,there was no significant difference between the Guizhi plus Gui dosage groups(P>0.05);(2)High-density lipoprotein and low-density lipoprotein: compared with the model group,metformin group,Guizhijia The high-density lipoprotein in the high-dose group of Guizhong was significantly increased,and the low-density lipoprotein was significantly decreased(P<0.05).Compared with the metformin group,the high-density lipoprotein increased significantly in the high-dose group of Guizhi plus Gui.The difference was statistically significant(P<0.05).There was no significant difference between the low-density lipoprotein and the Guizhi plus guinea group(P>0.05).(3)Myocardial zymogram: Compared with the model group,the metformin group,The high dose group of Guizhi plus Gui was significantly relieved,the difference was statistically significant(P<0.05).Compared with the metformin group,there was no significant difference between the Guizhi plus Gui dosage group(P>0.05).3.Histopathological changes of heart tissue: no pathological changes occurred in the blank group,the heart capsule was intact,the contours of the ventricle and ventricular muscle were clear,the myocardial fibers were arranged regularly and tightly;the fibroblasts appeared in the model group,and the myocardial cells were degenerated and necrotic;Compared with the model group,the dose of Zhijiagui granules and the metformin group were lighter.4.Changes in oxidative stress related indicators:(1)SOD,NOS,GSH-PX,MDA: Compared with the model group,the activity of SOD,NOS and GSH-PX in the high dose group of metformin group and Guizhi plus Guizhong was significantly increased.The content of MDA decreased significantly,and the difference was statistically significant(P<0.05).Compared with the metformin group,there was no significant difference between the different doses of Guizhi plus Gui(P>0.05).(2)Relative expression of NOX4 protein: and model Compared with the group,the expression of NOX4 in the metformin group and the Guizhi plus Gui high dose group was significantly lower(P<0.05).Compared with the metformin group,there was no statistical difference between the Guizhi plus Gui dosage group.>0.05);(3)Relative expression of p47 PHOX protein: Compared with the model group,the expression of p47 PHOX was significantly lower in the metformin group,Guizhi plus Guizhong high-dose group,the difference was statistically significant(P<0.05),compared with the metformin group.There was no significant difference in the dosage of Guizhi plus Gui(P>0.05).Conclusion:1.The blood glucose,blood lipids,glycosylated hemoglobin and myocardial enzymes of the model group were higher than other groups after treatment,and the myocardial pathological findings were the most obvious,indicating successful modeling.2.Guizhi Jiagui Granule can reduce myocardial damage and reduce the degree of myocardial necrosis and fibrosis in diabetic rats through Wenyang Yiqi and TiaoHeyingwei.3.Guigui Jiagui Granule can increase the activity of SOD,NOS and GSH-PX and decrease the content of MDA and the expression of NOX4 and p47 PHOX,indicating that Guizhi Jiagui Granule can inhibit myocardial oxidative stress to reduce myocardial damage.
