Objective: To investigate the effect of acupuncture of Baihui-Qubin on the expression of P75 NTR in the brain tissue of rats with acute cerebral hemorrhage,and to explore the mechanism of the acupuncture therapy for acute cerebral hemorrhage.Methods: 72 healthy adult male Wistar rats with body mass(300 ± 20)g were selected and randomly divided into sham operation group,model group and acupuncture group.Each group was divided into four subgroups according to different time points at 6h,24 h,3d and7 d,with 6 rats in each subgroup.The rat model of cerebral hemorrhage was prepared according to the modified autologous blood injection method.The rats in the acupuncture group were given the acupuncture of Baihui-Qubin.The Rosenberg scoring method was used to comprehensively evaluate the neurological deficits of the rats in each group.TUNEL assay was used to detect the apoptosis of perihematoma cells in the brain tissue of rats in each group,and immunohistochemistry was used to detect the expression level of P75 NTR in perihematoma of brain tissue,and the correlation between them was analyzed.Results:1.Results of neurological deficit scores:(1)No signs of neurological deficits were observed in the sham operation group;(2)Compared with the sham operation group at the same time,the rats in the model group showed obvious signs of neurological impairment.(P<0.01);(3)The rats in the model group showed signs of impaired neurological function at6 h after surgery,which was the most serious signs at 3d and improved at 7d;(4)The signs of neurological deficits in the acupuncture group were significantly improved,compared with the model group at the same time,the difference was statistically significant(P<0.05).2.TUNEL results:(1)There was a small amount of apoptosisin rats in the sham operation group;(2)Compared with the sham operation group at the same time,the model group and the acupuncture group had significantly different degrees of apoptosis in the rat brain tissue(P<0.01);(3)The apoptotic cells of the model group began to increase at 6h after operation,and the number of apoptotic cells was the highest at 3d,and decreased at 7d;(4)Compared with the model group at the same time,the number of apoptosis at different time points in the acupuncture group of rats were decreased,the difference has statistical significance(P<0.05).3.Immunohistochemical test results:(1)There were a small number of P75 NTR positive cells in the brain tissue of rats in the sham operation group;(2)Compared with the sham operation group at the same time,the number of P75 NTR positive cells in the brain tissues of the model group and the acupuncture group was significantly increased,with a statistically significant difference(P<0.05);(3)The P75 NTR positive cells in the rat brain tissue of the model group appeared 6h after surgery,and reached a peak at 3d,and began to decline at 7d;(4)Compared with the model group at the same time,the number of P75 NTR positive cells in each time point of acupuncture group were significantly reduced,the difference has statistical significance(P<0.05).4.Correlation analysis results: The expression of P75 NTR was positively correlated with the number of apoptosis,and the correlation coefficient was 0.811.Conclusion:1.Acupuncture of Baihui-Qubin can alleviate the signs of neurological deficits in rats with cerebral hemorrhage;2.Acupuncture of Baihui-Qubin can reduce the apoptosis of nerve cells after cerebral hemorrhage;3.Acupuncture of Baihui-Qubin can inhibit the expression of P75 NTR which is closely related to apoptosis,and has a protective effect on the brain tissue in the penumbra zone of cerebral hemorrhage.
