| Background Diabetic Nephropathy(DN)is one of the major causes of Chronic Kidney Disease(CKD)in China,has not been effective treated.The Traditional Chinese Medicine Panax Notoginseng and Panax Notoginseng Saponins have been used for clinical prevention and treatment of DN,but the specific mechanism of action and active ingredients has not been fully elucidated,therefore,become the bottleneck of limiting its clinical widely use.In order to overcome this problem,this project uses the DN animal model to further explore the mechanism and its effective components of prevention and treatment of DN.Objectives To explore the role and molecular mechanism of Panax Notoginseng Saponins and their effective components of Notoginsenoside Fc in the prevention and treatment of DN Methods In this study,continuous intravenous administration of Panax Notoginseng Saponins in STZ-induced diabetic SD rats and continuous intragastric administration of Notoginsenoside Fc in db/db diabetic mice.During this period,biochemical indexes of urinary protein and other animal models were examined.Changes in renal ultrastructure and pathological changes were observed.The changes of renal podocyte adhesion molecules and the expression of oxidative stress and apoptosis were examined.The level of PTEN/PDK1/Akt/mTOR pathway were examined.Panax Notoginseng Saponins and Notoginsenoside Fc have protective effects on DN renal podocytes through their related mechanisms.Results Panax Notoginseng Saponins and Notoginsenoside Fc can significantly reduce urinary protein in model animals,and significantly improve the pathological changes of kidney morphology,reduce podocyte injury,and delay the progress of DN through PTEN/PDK1/Akt/mTOR pathway to protect the kidney.Conclusion Panax Notoginseng Saponins and Notoginsenoside Fc can improve renal podocyte oxidative stress and apoptosis,delay DN progression and alleviate renal podocyte injury through PTEN/PDK1/Akt/mTOR pathway. |
