Preparation of CUR-m PEG-PLGA Nanoparticles by Modified-self-emulsion Solvent Evaporation MethodObjective: To prepare curcumin loaded Methoxypolyethylene-glycols-poly(lactic-coglycolicacid)(m PEG-PLGA)Nanopaticles,investigate their pharmaceutical characters.Methods: Use poly(ethylene glycol)-poly(s-caprolactone)as the carrier,TPGS as the surfactants,prepare curcuminloaded m PEG-PLGA nanopaticlesby emulsion solvent evaporation.Based on the single factorexperiment was adopted to optimized formulation and technology ofthe preparation of CUR-NPs.Morphology of CUR-NPs prepared bybest technology were observed by transmission electronmicroscope.Mean particle size,PDI and Zeta potential were estimated by laser particle size analyzer.Entrapment efficiency and drug loading were investigated by the method of centrifugation and high-performance liquid chromatography(HPLC).The drug release behavior of CUR-NPs in p H 5.8 PBS with 20% ethanol was studied by dialysis method.Results:The morphology of CUR-NPs was spherical.The mean particle size,Zeta potential,entrapment efficiency and drug loading were(376.3±2.69)nm,(-29.4±0.110)m V,(78.33±2.52)% and(7.76±0.51)%,respectively.The results indicated that the CUR-NPs suspension eliminated slower and presented a prolonged release and have a good water solubility.Conclusion: CUR-NPs prepared by emulsion solvent evaporation method was smooth and round with well-distributed particle size as well as high entrapment efficiency and drug loading.CUR-NPs showed the obvious sustained-release characteristic.Part ? Action of curcumin on Reversing Corticosteroid Resistance Induced by Cigarette Smoke Extract and Comparison of Biological Activity between curcumin and CUR-NPs NanoparticlesObjective: To investigate the effect of curcumin on reversing corticosteroid resistance induced by cigarette smoke extract and comparison of biological characteristics between curcumin and CUR-NPs nanoparticles.Methods: Cellular uptake of curcumin and CUR-NPs in RAW264.7 macrophages and RTE cells was determined by cellular fluorescence intensity observed and detected by laser confocal microscopy imaging.Cigarettesmoke extract stimulated RAW264.7 cell line and RTE cells to induce the processe of corticosteroid resistance in vitro.curcumin,dexamethasone,curcumin+ dexamethasone and CUR-NPs+dexamethasone were used to treat macrophage cell line RAW264.7 and RTE cells activated by cigarette smoke extract.The protein levels of IL-8 were measrured by ELISA.The protein levels of HDAC2 were measured by western blot.The m RNA levels of HDAC2 were measured by Realtime PCR.Results: Cellular uptake of CUR-NPs in RAW264.7 macrophages was higher than the curcumin.Compared with the groups of cigarette smoke extract,curcumin and budesonideesonide,the protein levels of IL-8 are decreased in cell of the curcumin +budesonide and CUR-NPs+ budesonide groups,the protein levels of IL-8 of CUR-NPs+ budesonide group is lower than curcumin+budesonide group.Compared with the groups of cigarette smoke extract and budesonide,the m RNA and protein levels of HDAC2 of curcumin and CUR-NPs groups is higher,the level of CUR-NPs group is higher than curcumin group.Conclusion: curcumin could improve HDAC2 expression levels to reversing the corticosteroid resistance induced by CSE.CUR-NPs nanoparticles have more biological activity than curcumin.
