Background:The etiology of idiopathic pulmonary fibrosis?idiopathic pulmonary fibrosis,IPF?is unclear.In recent years,it has been found that?gastroesophageal reflux disease,GERD?may play a role in the pathogenesis of IPF.A retrospective study found that anti-acid therapy is associated with prolonged survival in patients with IPF,but there is still a lack of prospective studies to confirm the importance of anti-acid in IPF treatment.Objective:To study the effect of anti-acid therapy on pulmonary fibrosis rats.Methods:Thirty-six healthy male-related SD rats were randomly divided into normal control group?N group,n=10?,model group?M group,n=6?,acid inhibition group?A group,n=10?and hormone treatment group?B group,n=10?.M,A.In group B,bleomycin-induced pulmonary fibrosis was induced by nasal drip,while group N was intranasally injected with normal saline.On the 7th and 14th day after the establishment of the model,one rat in group M was killed at random,and the remaining four rats in group M were killed on the 28th day after the establishment of the model to verify whether the model was successful or not.Lung tissue was examined by HE staining and serum malondialdehyde?malondialdehyde,MDA?was detected to evaluate the degree of cell injury.Arterial blood gas analysis of oxygen partial pressure?Partial Pressure of Oxygen,PaO2?was used to evaluate the changes of pulmonary function in rats.After the establishment of the model,ranitidine hydrochloride capsule,prednisone acetate tablet and saline were used to treat group A,B,N by gastric perfusion every day.After 28 days of treatment,all rats were killed for the above determination.Results:The degree of alveolitis and fibrosis in group A and B were significantly lower than those in group M?P>0.05?,and there was no significant difference between group A and group B?P>0.05?.The content of MDA in group M was significantly higher than that in group N?t=15.93,P<0.01?.The content of MDA in group A was significantly lower than that in group M?t=4.79,P<0.01?.The content of MDA in group B was significantly lower than that in group M?t=6.20,P<0.01?.Compared with group B,there was no significant difference in MDA content between group A and group B?P>0.05?.The Pa02 value of blood gas analysis was not significantly different between group B and group M after treatment?P>0.05?,and there was no significant difference between group B and group M after treatment?P>0.05?.Conclusion:To some extent,the anti-acid therapy can improve alveolar inflammation and pulmonary fibrosis in rats with pulmonary fibrosis,and it has similar effect as prednisone acetate in improving pulmonary fibrosis.
