Objective1)This study aimed to observe the effect of Zuogui Wan,a Chinese herbal compound for tonifying kidney,on the three-dimensional architecture of the reproductive organ vessels in perimenopausal rats.2)To investigate the effects of Bushen Chineses medicine Zuogui Wan on circulating endothelial progenitor cells and its mobilization factors of E2 and VEGF in perimenopausal rats..Methods1)Thirty-two perimenopausal rats.aged 10-12 months were randomly divided into four groups:model control group?M?,positive medicine control group?CP?,low-dose Zuogui Wan group?LD?,and high-dose Zuogui Wan group?HD?,with 8 rats in each group.Another 8 female rats aged 4-6 months were selected as black control group?B?.After 15 days of gavage treatment,Three-dimensional architecture of reproductive organ vessels in rats were quantitatively analyzed by vasucular casting and scanning electron microscopy.2)Forty perimenopausal rats.aged 10-12 months were randomly divided into four groups:model control group?M?,positive medicine control group?CP?,low-dose Zuogui Wan group?LD?,and high-dose Zuogui Wan group?HD?,with 10 rats in each group.Another 10 female rats aged 4-6months were selected as black control group?B?.After 15 days of gavage treatment,The changes of ovarian morphological were observed,and the organ index of ovary and uterus was calculated.The proportion of CD34+/CD133+/VEGFR2+EPCs in peripheral blood mononuclear cells was detected by flow cytometry.The level of mobilization factors E2 and VEGF was detected by ELISA.Results1)Study showed that follicles at different developmental stages,corpus luteal,dense vessel networks on uterus,and vagina with a large number of spiral vessels in rats were observed?under scanning electron microscopy?.The vessel number,inside diameter,and area percentage of ovary,uterus,and vagina were increased in high-dose Zuogui Wan group compared with those in model control group?P<0.05 or P<0.01?.2)Zuogui Wan 33 g/kg group could increase the number of follicles,corpus luteum,blood vessels?P<0.05 or P<0.01?,and raise the level of E2,VEGF and EPCs?P<0.01?.Conclusion1)The results show that Zuogui Wan may promote angiogenesis of reproductive organ and improve three-dimensional architecture of reproductive organ microvessels in perimenopausal rats.2)Zuogui Wan might enhance the level of E2 and VEGF,and mobilize EPCs,thereby promote ovarian angiogenesis and improve ovarian function.
