Aims: Vascular remodeling and VSMCs phenotype switch are the foundation of many kinds of cardiovascular disease,just like atherosclerosis,restenosis,hypertension and so on.This study aims to explore the role of MBNL1 in the regulation of vascular remodeling and VSMCs phenotype switch.Method: establish model of rat carotid artery injury,then HE staining is used to check whether the neointima is formation,western blot is used to detect the expression of MBNL1 in the artery.Treat the rat VSMCs with PDGF-bb,then detect the the expression of MBNL1 with different treating time and different dose of PDGF-bb.Overexpress MBNL1 with the lentivirus and knockdown it with siRNA,western blot is used to detect the expression of VSMCs marker genes,transwell is used to detect the migration ability,Edu is used to detect the proliferation.Result: The expression of MBNL1 in the injury artery is higher than in the normal one.Treating the VSMCs with PDGF-bb can increase the expression of MBNL1.Overexpression of MBNL1 enhances the the expression of marker genes as well as inhibiting the migration of VSMCs.Knockdown of MBNL1 has opposite effect.Neither overexpression nor knockdown of MBNL1 affects the ability of proliferation.Conclusion: The expression of MBNL1 increases in injury artery and VSMCs treated with PDGF-bb.MBNL1 can up-regulate the expression of marker genes,and depress the migration of VSMCs.MBNL1 has no effect on proliferation.
