Objective: to detect the expression of glucose regulation protein 78(GRP78)in peripheral blood serum and placental trophoblast cells of normal pregnant women and preeclampsia patients,and to explore its relationship with the pathogenesis of preeclampsia.To determine whether it can be used as a biomarker for the prediction of preeclampsia and provide a basis for the early diagnosis of preeclampsia.Methods:1.The object of study: according to the gestational hypertension disease diagnosis and treatment guidelines(2015)in the diagnosis of preeclampsia,choose in May 2017 to May2018 in Qingdao municipal hospital eastern hospital maternity center line termination of gestation preeclampsia pregnant women in cesarean section 60 cases for treatment group,30 cases of early-onset and late-onset 30 cases,swap over the same period of 30 cases of normal pregnancy pregnant women as control group.2.Specimen collection:the extraction of pregnant women before delivery elbow venous blood 2 ml,4 ? 1000 r/min,the centrifugal clear liquid on 15 minutes after,at-80 ?refrigerator waiting for inspection;The size of placenta tissue in the whole layer near the central umbilical cord root(avoiding necrosis,hemorrhage and calcification)was about2.0cm x 1.0cm x 1.0cm immediately after delivery of the placenta.After washing with normal saline,sterile gauze was used to absorb water and placed into a liquid nitrogen tank for freezing for inspection.3.Test indicators: double antibody sandwich ELISA was used to detect GRP78 content in serum.The contents of GRP78 protein and m RNA in placental trophoblast cells were determined by Western blotting and semi-quantitative reverse transcriptase polymerase chain reaction(rt-pcr).4.Data analysis: SPSS 23.0 software package was used for statistical analysis.Results:1.The expression level of GRP78 in serum of each group was 0.3 0.02 in the early onset group,0.26±0.02 in the late onset group,and 0.19±0.15 in the normal pregnant women group.GRP78 protein content in serum of each group of preeclampsia was higher than that of the normal pregnant women group,and the difference was statistically significant(P<0.05).There was no significant difference between the groups(P>0.05).2.GRP78 protein expression levels in placental tissues of each group: 0.74±0.16 in the early onset group,0.75±0.19 in the late onset group,and 0.41 0.04 in the normal pregnant women group.GRP78 protein content in serum of each group of preeclampsia was higher than that of the normal pregnant women group,and the difference was statistically significant(P<0.05).There was no significant difference between the groups(P>0.05).3.GRP78 m RNA expression levels in placental tissues of each group were 2.53 ±0.03 in the early onset group,2.59±0.05 in the late onset group,and 1.15 0.04 in the normal pregnant women group.GRP78 protein content in serum of each group of preeclampsia was higher than that of the normal pregnant women group,and the difference was statistically significant(P<0.05).There was no significant difference between the groups(P>0.05).Conclusion: GRP78 expression is increased in serum and placental trophoblast cells of preeclampsia patients,suggesting that changes in GRP78 expression may be related to the pathogenesis of preeclampsia.
