Objective: 1.Detect the expressions of lnc RNA-UCA1,HMGB1,RAGE,NF-?B p65 protein and m RNA in human NSCLC and adjacent normal tissues.2.Through the expression correlation of lnc RNA-UCA1,HMGB1,RAGE,NF-?B p65 in human NSCLC,it is speculated that lnc RNA-UCA1 works on NF-?B pathway through HMGB1 / RAGE signal axis,thus affect the occurrence of NSCLC.3.To investigate the expression of lnc RNA-UCA1,HMGB1,RAGE and NF-?B p65 in NSCLC,and the relationship between clinical characteristics and pathological types of lung cancer.Methods: 1.Collect 40 fresh specimens from the department of thoracic surgery of Affiliated hospital of Qingdao University from May 2018 to August 2018,all cases were confirmed by pathological diagnosis as NSCLC,and take the corresponding normal tissues adjacent to the tumor?5cm from the tumor boundaries,no tumor cell infiltration was confirmed by pathological diagnosis?.2.Immunohistochemical method was applied to observe the staining of HMGB1 and RAGE in cancer tissues and adjacent normal tissues of 10 patients with NSCLC,and to analyze their localization and expression quantity in cells.3.Western blot semi-quantitative protein detection was used to detect the expression of HMGB1,RAGE and NF-?B p65 protein in cancer tissues and adjacent normal tissues of 12 patients with NSCLC.4.Real-time quantitative gene amplification fluorescence detection?q PCR?was used to detect the m RNA expression of lnc RNA-UCA1,HMGB1,RAGE and NF-?B p65 in cancer tissues and adjacent normal tissues of 40 patients with NSCLC.The internal reference was ?-actin,and calculated the relative expression quantity of the above indicators in NSCLC.The results were expressed by 2-??Ct.Results: 1.The expression of HMGB1 and RAGE was detected by immunohistochemical method.The results shows that the positive staining area of HMGB1 was mainly for the nucleus.The positive staining area of RAGE was mainly in the cytoplasm of cells.The staining area of HMGB1 in cancer tissues is larger than that of normal tissues,while RAGE in cancer tissues is smaller than that of normal tissues,indicating that HMGB1 is highly expressed in NSCLC,while RAGE is low.2.The expression of HMGB1,RAGE and NF-?B p65 in lung cancer tissues and adjacent normal tissues of 12 patients with NSCLC were detected by Western blot.The results shows that the expression of HMGB1,NF-?B p65 is significantly higher than adjacent normal tissues,while RAGE is lower than adjacent normal tissues.3.The q PCR method was used to detect the expression quantity of lnc RNA-UCA1,HMGB1 m RNA,RAGE m RNA and NF-?B p65 m RNA in lung cancer tissues and adjacent tissues of 40 patients.The results shows that in NSCLC,the expression of HMGB1 m RNA and NF-?B p65 m RNA was significantly higher than that of normal tissues,while the expression of RAGE m RNA was significantly lower than that of normal tissues.4.In NSCLC,the expression level of lnc RNA-UCA1 is highly positively correlated with HMGB1,and the expression level between HMGB1 and RAGE is highly negatively correlated,and the expression level between RAGE and NF-?B p65 is highly negatively correlated.The expression level was highly negatively correlated between HMGB1 and NF-?B p65.5.The expression levels of lnc RNA-UCA1,HMGB1,RAGE,and NF-?B p65 are closely related to the stage,differentiation,tumor size,lymph node metastasis and pathological type,the expression of lnc RNA-UCA1,HMGB1 and NF-?B p65 at stage II and III was significantly higher than that of stage I,and was significantly higher than that of low stage when poorly differentiated.The large tumor was significantly higher than the tumor of small volume,and lymph node metastasis was significantly higher than the absence of lymph node metastasis,the expression in squamous cell carcinoma was significantly higher than that in adenocarcinoma;the expression of RAGE was exactly the opposite,significantly lower in stage II and III than in stage I,and significantly lower in poor differentiation than in poor differentiation.The small tumor was significantly higher than the tumor of large volume,lymph node metastasis was significantly lower than that without lymph node metastasis,and the expression level in squamous cell carcinoma was significantly lower than that in adenocarcinoma;lung cancer patients with different ages and smoking history had no effect on the expression levels of lnc RNA-UCA1,RAGE,HMGB1,and NF-?B p65.Conclusion: 1.lnc RNA-UCA1,HMGB1,NF-?B p65 is highly expressed in NSCLC,and RAGE is low.2.The expression of lnc RNA-UCA1,HMGB1,RAGE,and NF-?B p65 is correlated in NSCLC.It is speculated that lnc RNA-UCA1 can work on NF-?B pathway through HMGB1/RAGE signal axis,and has a certain influence on the occurrence and development of NSCLC.3.The expression of lnc RNA-UCA1,HMGB1,RAGE,NF-?B p65 is closely related to tumor stage,tumor size,tumor differentiation,pathological type and lymph node metastasis.It is further proved that the above indicators are related to tumor metastasis and invasion.
