Objective:The occurrence of prostate cancer with racial diversity,is studied in this article,through the transcriptome sequencing of 3 cases of Mongolian men with prostate cancer gene expression profile,and its using bioinformatics sequence and the han nationality and Mongolian normal prostate cancer patients are compared,and the gene expression profile in order to get the Mongolian dysregulated prostate cancer patients,the Mongolian prostate cancer susceptibility genes to lay the experimental foundation for mining.Methods:Three Mongolian prostate cancer patients were divided into experimental group(T group,including T1,T2 and T3 groups).Han prostate cancer patients were divided into positive control group(T4 group);Mongolian normal people were divided into the normal control group(C1 group).The blood of patients and normal people in the above groups were collected and RNA was extracted.Illumina sequencing platform was used for transcriptome sequencing.Gene expression levels of each group were compared.GO functional annotation and GO enrichment analysis were performed with bioinformatics software for significantly different genes,and specific expression genes of Mongolian prostate cancer patients in Inner Mongolia were obtained through comparative analysis.RESULTS:Through high-throughput RNA sequencing and bioinformatics analysis,the following results were obtained from the comparison between the groups:1.Analysis results of gene differential expression between the experimental group and the positive control group:①.In comparison with T4 gene expression profile,there were 3046 up-regulated genes and 875 down-regulated genes.Among them,18 differentially expressed genes were significantly up-regulated,including ELANE、SIGLEC8、DIRAS1、KLF2、OLFM4、CBLN3、TSPO、CRISP3、OLFM4、LCN2、CDH2、TMPO、PGLYRP1、RETN、CD74、LY6E、SELENBP1、CDH2,and 9 differentially expressed genes were significantly down-regulated.Including CXCR1、AQP9、MT-ND6、MT-C02、MT-ATP6、MT-ND2、MT-CYB、MT-RNR1、PI3②.The T2 and T4 gene expression profile were compared,with 440 up-regulated genes and 6694 down-regulated genes.Among them,13 genes were significantly up-regulated,including ELANE、HBM、FTH1、PPDPF、HBA1、CRISP3、OLFM4、LCN2、SELENBP1、LY6E、TSPO、CD74、RETN,and 9 genes were significantly down-regulated,including MT-ND6、MT-C02、MT-ATP6、MT-ND2、MT-C02、AQP9、CXCR1、LYN、MCL1.③.Comparison of T3 and T4 gene expression profiles showed that 6745 genes were up-regulated and 430 genes were down-regulated.Among them,13 genes were significantly up-regulated,including CRISP3、TRBJ1-4、EEF1A1P5、RPL41、OLFM4、LCN2、S1PR1、HSP90B1、PGLYRP1、RETN、APEX1、LY6E、ZAP70,and 8 genes were significantly down-regulated,including ADIPOR1、MT-ND6、MT-C02、MT-ATP6、MT-ND2、AQP9、CXCR1、MT-CYB.④ Comparison of gene expression profile between T group and T4 groupCompared with han patients with prostate cancer transcriptome,three Mongolian of genes expressed in prostate cancer patients raised together 10,including CRISP3,LCN2,high ELANE,TSPO gene expression related to cell invasion,high SELENBP1 gene expression related to cell proliferation,KLF2,high OLFM4 gene expression related to the inhibition of tumor and CD74,androgen dependence in prostate cancer),high RETN gene expression related to cell proliferation,high LY6E gene expression related to promote angiogenesis and tumor cell proliferation.Among them,the low expression of AQP9 gene is related to apoptosis,the low expression of CXCR1 gene is related to cell proliferation,MT-ND6、MT-C02、MT-ATP6、MT-ND2are mitochondrial genes,and their low expression is related to the occurrence and development of PCa.After further GO functional analysis,CXCR1 gene was also related to cell proliferation.KLF2 gene expression is related to hif-1/Notch-1 signaling pathway.2.Analysis results of gene differential expression between the experimental group and the normal control group①The expression profiles of T1 and C1 genes were compared.There were 2,104 up-regulated genes and 2,122 down-regulated genes.Among them,17 genes were significantly up-regulated,including IFI27,OAS1,ISG15,OLFM4,PGLYRP1,GPX1P1,LY6E,CRISP3,MMP-9,OLFM4,ARG1,RETN,CD24,LCN2,LY6E,TSPO and MPO,and 18 genes were significantly down-regulated,including CCL5、ALPL、NAMPT、MT-COI、MT-ND2 MT-ND5、MT-ND1、MT-C03、JUNG、CD74、MT-ATP6、MT-C02、MT-ND6、MT-CYB、MT-ND4 MT-RNR1、MT-ND3、TXNIP.②The expression profiles of T2 and C1 were compared,among which 450 were up-regulated and 11,408 were down-regulated,among which 15 were significantly up-regulated,including CRISP3,OLFM4,SELENBP1,MMP-9,ISG15,PDZK1IP1,LY6E,PHOSPHO1,RETN,ADIPORI,LCN2,MPO,CD24,OAS1 and LY6E,and 12 genes were significantly down-regulated,including CD 14、CCL5、ALPL、NN2、ARF1、RACK 1、JUNB、NAMPT、MT-ND6、MT-C02、MT-ATP6、MT-ND2.③Compared with C1 gene expression profile,2872 genes were up-regulated and 630 genes were down-regulated.Among them,23 genes were significantly up-regulated,including CRISP3,RPS7P1,RPL23,TRBJ1-6,RPL7,GZMA,OLFM4,LY6E,SELENBP1,ISG15,IGLC3,ARG1,PGLYRP1,PDZK1IP1,MMP-9,RETN,LCN2,PDZK1IP1,LY6E,MPO,CD24,OAS1 and TSPO.15 genes were significantly down-regulated,including CXCR1、CCL5、ALPL、ADIPOR1、NAMPT、MT-RNR1、JUNB、MT-ND6、MT-C02、IFIT2、R3HDM4、HBA2、MT-ATP6、HBB、MT-ND2.④Gene expression profiles of group T and C1 were comparedThree Mongolian men with prostate cancer compared with control group,jointly increase the expression of genes in 11,including CRISP3,high LCN2,MMP-9 gene expression related to cell invasion,OLFM4,high PGLYRP1 gene expression related to the inhibition of tumor,high LY6E gene expression to promote angiogenesis and tumor cell growth,high RETN,ARG1 gene expression related to regulate the cell cycle and cell proliferation,MPO,CD24,OAS1 gene associated with cancer,the exact mechanism is unclear.Compared with control group,three Mongolian men with prostate cancer common down expression of genes have eight,including CCL5,low ALPL gene expression related to cell invasion,low NAMPT gene expression related to cell growth,low JUNB gene expression related to cell proliferation,MT-ND6、MT-C02、MT-ATP6、MT-ND2 is mitochondrial genes,its development in low expression and PCa.Further GO functional analysis revealed that CCL5 and ARG1 gene expression levels were also related to the immune response of the body,JUNB gene expression was related to cell proliferation,and RETN and LY6E were related to cell pathwaysConclusion:①After transcriptome sequencing and bioinformatics analysis,there were 16 significantly differentially expressed genes in patients with prostate cancer in Mongolia and han,among which10 genes were significantly up-regulated and 6 genes were significantly down-regulated.②compared with the normal Mongolian patients with prostate cancer,there are 19 significantly differentially expressed genes,including 11 genes significantly up-regulated and 8 genes significantly down-regulated.We speculate that these specific genes expressed in Mongolian prostate cancer patients may be associated with the occurrence of Mongolian prostate cancer.
