| Alzheimer’s disease(AD)is a one of the most prevalent neurodegenerative diseases.There are 4 people diagnosed with AD per second and almost 440 million people in the world.AD is belong a complex disease and induced by the various factors.Although researches efforts on the pathological characteristics and biological metabolism and regulation in the past year,but the cause and mechanism of AD was remain unclear.Currently,increasing evidences indicated that the gut microbe has been implicated in metabolism,immunity,and health maintenance of its host especially in the host immune system and the nervous system.In this study,we carried out the analysis and metagenome sequencing used by the APP/PS1 transgenic mice as AD-prone model.Our results showed as follows:(1)There were no significant difference between AD-prone group and wild-type group at 3 months old,but significantly differently at 6 and 9 months old on the memory cognitive ability.In other hand,more disposition of β-amyloid protein was detected in AD-prone mice by withimmunohistochemistry test.It indicated the AD-prone model was successful and useful for next works.(2)The diversity of gut microbiota reduced along the progress of AD.(3)The composition of gut microbiota influence dremarkably by with the host genotype and age.At the phylum level,4 dominant phylumfrom among 26 phylum were indicated significant difference,such as Actinobacteria,Bacteroidetes,Firmicutes and Proteobacteria in every stage.At the genus level,the aboundance of some bacterials among 243 genus were dynamic changed between the different genotype and ages,such as the Lactobacillus,Hyphomicrobium,Mucispirillum,Anaerococcus,Hydrogenophilus and Thermus.(4)After feeding with a probiotic of L.casei Zhang,the gut microbiota of the AD-prone mice and wild-type mice changed,respectively.(5)The abundance of some bacterial communitys also remarkably influenced by the gender factor such as Bifidobacterium,Pseudonocardia and Acetobacter.(6)Further,with diffent gut microbes between the AD-prone mice and wild-type mice,the microbial function prediction indicated that the function pathways were main enrichmented to the immune system disea,neurodegenerative diseases,cell growth and death,Carbohydrate synthesis and metabolism.We infer the symbiotic interactions between host AD-prone mice and gut microbes was through by with the systemof microbiota-gut-vagus-brain axis. |
