Research Of Chemiluminescent Immunoassay For Tumor Marker CA242 In Serum

Hunman serum tumor marker carbohydrate antigen 242(CA242)is a type of salivary,mucus-type carbohydrate antigen.As a commonly used clinical trial,when the body's normal tissues is cancerous,the important component of the cell membrane glycoprotein or glycolipid will change.Therefore,CA242 is used for the screening,detection and diagnosis of the tumor.This study aims to estabilish a magnetic particle chemiluminescent immunoassay for the quantitative measurement of serum tumor marker carbohydrate CA242 assay kits.We choose one CA242 antibody coating with magnetic particle,another antibody labeling with Alkaline Phosphatase(ALP),the immunoassay “Double Antibody Sandwich” method was employed to detect CA242 in the sample.The immune complexes,which combined with CA242 antigen and CA242 antibody,were captured with magnetic particle.The chemiluminescent substrate was added to react with ALP labeled with antibody.The concentration of CA242 can be automatically calculated by the instrument.Analysis of performance evaluation showed that the assay precision between 85% and 115%,.The assay was linear between 1 to 150U/mL and had an assay limit detection of the assay is calculated to be lower than 1U/mL.Repeatability as evaluated from the within-run CV was less than 8% and between-run CV was less than 15%.In addition,the test results showed that there was no interference when serum have bilirubin,hemoglobulin,2% intralipid,rheumatoid factor and 160 titre ANA were contained in the samples.Finally the clinical sample results between our own and the listed assays kit showed good comparability and comparison agreement.In conclusion,the CA242 quantitative assay established in this study had good performance in sensitivity,anti-interference and precision.Our reagents can be used in the diagnosis of malignant tumor.
