| Objective:To explore the protective effect of curcumin on traumatic brain injury in rats and to explore its potential mechanism.Methods:The healthy male SD rats were randomly divided into four groups:sham group,TBI group,TBI+curcumin 20 mg/kg group,TBI+curcumin 200mg/kg group.The neurological deficits were scored in each group and compared with the brain water content and the permeability of the blood-brain barrier after 24h.The neuronal apoptosis was detected by TUNEL,and the levels of oxidative stress and Ca2+in brain tissue were measured.The expression levels of Bcl-2,Bax and Caspase-3 were detected by Western Blot.Results:The neurological deficit score showed that curcumin significantly improved neurological function in rats with traumatic brain injury and significantly reduced brain water content and permeability of the blood-brain barrier.TUNEL experiments show that curcumin can significantly inhibit the apoptosis of neuronal cells,relieve oxidative stress in brain tissue,This protective effect may be achieved by reducing the concentration of Ca2+in the brain tissue.Curcumin can inhibit endogenous apoptosis signaling pathway and reduce caspase-3 expression.Conclusions:Curcumin has a protective effect on the neurological function of rats with traumatic brain injury,it can reduce the neuronal cell apoptosis and oxidative stress levels in brain tissue,and significantly inhibit the endogenous apoptosis signaling pathway.Which can provide some reference for future clinical treatment for traumatic brain injury. |
