Objective: By establishing a COPD mouse model and studying the surface molecules of m DC(CD83),im DCs(CCR6),Th1 cytokines(IFN-?,IL-12)and Th2 cytokines(IL-4,IL-5)to investigate the changes and significances of CD83,CCR6,Th1 and Th2 related cytokines in mouse COPD model.Methods: The C57BL/6 mice of SPF grade were randomly divided into normal control group(n = 6)and COPD model group(n = 6).Exposure to cigarette-smoking was used to establish the mice model of COPD.Two groups of mice were killed and their bronchoalveolar lavage fluid(BALF)and lung tissue specimens were collected on the 28 th day of the experiment.Th1 and Th2 related cytokines(IFN-?,IL-12 and IL-4,IL-5)in lung tissue and BALF were detected by ELISA.Immunohistochemistry was used to detect CD83,CCR6,IFN-?,IL-4,CCR6/CD83 and IFN-?/IL-4 ratio in lung tissue.The histomorphology of lung tissue were observed by HE staining.Results:1.The lung tissue in the mouse COPD model group were stained by HE showed significant pulmonary inflammation and formation of emphysema.2.Compared with the control group,the expression of CD83 decreased and CCR6 increased in lung tissue of mouse COPD model,the CCR6/CD83 ratio increased,the differences were statistically significant(P<0.05).3.Compared with the control group,the expressions of Th1 cytokines(IFN-?,IL-12)increased and Th2 cytokines(IL-4,IL-5)decreased in lung tissues and BALF of mouse COPD model,at the same time,the IFN-?/IL-4 ratio increased,the differences were statistically significant(P < 0.05).4.There was a positive relationship between CCR6/CD83 and IFN-?/ IL-4 in lung tissue of mouse COPD model(r = 0.92).(P < 0.05).Conclusion:CD83,CCR6 and Th1,Th2 related cytokines are involved in the immune regulatory mechanism of mouse COPD model at the same time.
