Objective:To observe the different expression of SOCS1 mRNA,SOCS3mRNA,JAK2 mRNA,STAT3mRNA and phosphorylated protein in the decidua of placenta in threate ned abortion mice,and the expression of IL-6 in serum of pregnant mice in each grou p.To explore the mechanism whether the JAK2/STAT3/SOCS3 signaling pathway affect s the expression of active proteins in threatened abortion,and the mechanism of She ngshisan decoction in maintaining pregnancy.Methods:Forty female CBA/J mice were divided into 4 groups according to the r andom number table method.There were 4 groups in each group,which were normal pregnancy group,threatened abortion model control group,Shengshi Sanhua prescripti on high-dose treatment group,and corpus luteum.Ketone treatment group.Female and male mice were then caged at a ratio of 2:1.Ten female CBA/J mouse and five male BALB/c mouse were mated to form the normal pregnant model group,30 female CBA/J mouse and 15 male DBA/2 mouse were mated to form the threatened abortion mod el group.Use the method of random number table divided the threatened abortion mod el into three groups,which were the progesterone treatment group,the Shengshisan deco ction high-dose treatment group and thethreatened abortion model control group.The ex pression of SOCS1 mRNA,SOCS3mRNA,JAK2 mRNA and STAT3 mRNA were detected by RT-PCR.ELISA was used to detected the expression of IL-6 in serum of the pregn ant mice.Western blot was used to detect the expression of STAT3,p-STAT3,JAK2 and p-JAK2 proteins at the maternal and feta interfaces of pregnant mice in each group.Use the immunohistochemistry to detected the location of STAT3 and p-STAT3 in the maternal-fetal interface of pregnant mice.Results:1.RT-PCR results showed that Shengshisan decoction could increase SOCS3 mRNA and STAT3 mRNA and decrease SOCS1 mRNA.The results showed that SOCS1 mRNA,SOCS3mRNA,JAK2 mRNA and STAT3 mR NA were expressed in decidua and placental tissues of mice in each group,but comp ared with normal pregnancy group,SOCS1 mRNA was significantly increased in threaten ed abortion mice group(P<0.05),while the expression of SOCS3 mRNA and STAT3 mR NA was significantly decreased(P<0.05),and JAK2 mRNA was not significantly differe nt between groups(P>0.05).2.ELISA results suggested that Shengshisan decoction could increase the IL-6 ex pression in pregnant mice with threatened abortion.The results showed that IL-6 was expressed in decidua and placental tissue of mi ce in each group,but compared with normal pregnancy group,the expression of IL-6in pregnant mice in threatened abortion model control group was significantly lower(P<0.05),and the expression of IL-6 in Shengshisan decoction treatment group and prog esterone control group was not different from that in normal pregnancy group(P>0.05).3.Western blot results suggested that Shengshisan decoction could improve the e xpression of p-STAT3 at the maternal and fetal interface of pregnant mice.The results showed that: STAT3,p-STAT3,JAK2,p-JAK2 were expressed in pla centa and slough tissue of mice in each group: compared with the normal pregnancy group,p-STAT3 protein was significantly decreased in the threatened abortion model c ontrol group,have a statistical significance(P<0.05).The expression of p-STAT3 protein in the Shengshisan decoction and progesterone groups were similar as the normal pregnancy group,have no statistical significance(P>0.05).The expression levels of STAT3,JAK2 and p-JAK2 were not significantly different between the groups(P>0.05).4.Immunohistochemistry results that STAT3 is mainly expressed in the cell membrane and cytoplasm of trophoblast cells,while p-STAT3 is mainly expressed in the nucleus.The results showed that:STAT3 was mainly expressed in the cell membrane and cytoplasm of trophoblast cells,while p-STAT3 was mainly expressed in the nucleus.Compared with the normal pregnancy group,the expression of p-STAT3 in the maternal-fetal interface of the threatened abortion model control group was significantly lower(P<0.05).The expression of p-STAT3 protein in the high dose of Shengshisan decoction treatment group,and the progesterone treatment group was close to the normal pregnancy group,and there was no statistically significant(P>0.05).There was no significant difference in the expression of STAT3 between the model groups(P>0.05).Conclusion:1.Shengshisan decoction can increase the expression of SOCS3 mRNA andSTAT3mRN A at the maternal and fetal interface of pregnant mice,reduce the expression of SOCS1 mRNA,improve the immune response of Th network in pregnant mice,and maintain the high expression status of Th2 cytokines,thus maintaining pregnancy.This is consistent with the expression at the protein level.2.The expression of IL-6 in the body of the pregnant mice is affected by the shengshisan decoction,which can increase the expression level of IL-6,change the immune imbalance of Th1 high expression in pregnant mice with threatened abortion,and maintain the pregnancy.effect.3.Shengshisan decoction may promote the expression of STAT3 protein phosphorylation in the maternal-fetal interface of pregnant mice by stimulating the conduction of JAK2/STAT3 signaling pathway,so that the high level of p-STAT3 is maintained in the pregnant mother,and to maintain pregnancy.4.STAT3 is mainly expressed in the cell membrane and cytoplasm of trophoblast cells,while p-STAT3 is mainly localized in the nucleus.Shengshisan decoction can increase the expression of p-STAT3,stimulate the phosphorylation of STAT3,promote its nuclear translocation process,and maintain the pregnancy.
