Therapeutic Effect Of Hepatic Artery Infusion With P38MAPK Inhibitor On Hepatic Echinococcosis In Rats

Objective: To investigate the effect of hepatic artery infusion of p38 MAPK inhibitor SB-202190 reagent on hepatic hydatid rats.Methods:150 clean female Wistar rats weighing 250-300 g were purchased and provided by Xi'an Coo Biotechnology Co.,Ltd.for the establishment of rat hepatic hydatid model(confirmed by B-ultrasound three months later).Seventy-eight rats were successfully established,and then hepatic artery catheterization was performed.The remaining 67 rats were successfully established,with a total success rate of 44.7%.Sixty rat models of hepatic cystic echinococcosis were randomly selected and divided into three groups: saline control group(group A),50 micromol/L SB-202190(group B),100 micromol/L SB-202190(group C).Rats in groups A,B and C were perfused with 0.3 ml saline on 1d,3d,7d,14 d and 42 d respectively.Rats in group A were perfused with 0.3 ml of normal saline.Rats in group B were perfused with 0.3 ml of 50 micromol/L SB-202190 solution.Rats in group C were perfused with 0.3 ml of 100 micromol/L SB-202190 solution.After disinfection,rats were returned to cage and fed independently.Rats in each group drank water freely and ate the same diet.On the 43 rd day after the end of the administration,the peripheral venous blood of rats was collected to test the liver function and body weight of rats.Finally,all rats were killed.The liver tissues of rats were taken and fixed with 10% formaldehyde solution.The diameter of hepatic cystic echinococcosis was measured,and the pathological changes of hepatic echinococcosis in each group were observed under a high power microscope.Results: The liver function test results of rats on 43 rd day: The liver function test results of rats on the 43 rd day:(1)VS group A and VS group B VS group VS group VS group C group,ALT(49.58 +2.38 VS 38.35 +38.38 VS 38.35 +1.34 VS 30.35 3 3 3.35 +1.34 VS 30.93 +30.93 +1.51),AST(67.45 +5.14 5.14 VS 54.86.14 VS 54.86 16 VS 5.79(+0.15)VS 5.83(+0.31)P < 0.05 had statistical significance.(2)The weight of rats was measured on the 43 rd day.There was statistical significance in the weight of group A,group B and group C(0.248(+0.00695),0.273(+0.00957),0.282(+0.01465),P < 0.05.(3)On the 43 rd day,the diameter of rat hydatid internal capsule was measured.There was statistical significance in group A,group B,and group C(0.3830(+0.023)VS,0.2995(+0.0237)VS,0.2005(+0.0274),P < 0.05.(4)Observing the pathological sections of liver tissue under high power microscope,the infiltration of inflammatory cells in group C was lighter than that in group B and group A.Conclusion: SB-202190 inhibitor can improve the liver function of rats with hepatic echinococcosis,slow down the weight change of rats,reduce the diameter change of internal capsule and alleviate the inflammatory reaction of hepatic tissue,and inhibit the growth of hepatic echinococcosis in rats.
