Objective: The rat model of acute high altitude lung tissue injury was established by simulating the high altitude environment of low pressure and hypoxia.To observe the effect of bloodletting preconditioning on oxidative stress response of lung tissue injury in rats,which can provide new way to prevent and treat acute high altitude lung injury.Methods: Sixty-four adult male SD rats were randomly divided into control(A)group,bleeding pretreatment(B)group,24 h?48 h and 72 h of hypobaric hypoxia(C)group,24 h?48 h and 72 h of hypobaric hypoxia and Blood-letting Puncture on Twelve-well of Hand(D)group.Group B and D were pretreated with blood collection acupuncture according to the order of Shaoshang,Shangyang,Zhongchong,Guanchong and Shaoze for 5 days,and the amount of blood loss per point was 6 ? L,once a day;At the same time,rats in groups A and C were crawled once a day in the same way as rats in groups B and D.On the 6th day,the rats in groups A and B were taken,and the rats in groups C and D were placed in the simulated hypobaric oxygen chamber(7 000 m above sea level)to establish the model of acute high altitude lung injury.After the corresponding time of exposure in the cabin,the blood of abdominal aorta was taken out of the cabin,and the lung tissue was taken to observe the pathological changes of lung tissue,and dry-wet weight ratio,SOD,GSH activity,MDA and NO content of lung tissue in each group were measured.Results: 1.Compared with group A,the dry-wet weight ratio and MDA content in group C and D increased at each time point(P < 0.05),SOD and GSH activity decreased(P < 0.05),and NO activity increased first and then decreased(P < 0.05);pathology showed that the lung tissue structure of rats showed varying degrees of injury,and the degree of injury was serious at 72 hours.2.At the same time,the lung tissue indexes of the two groups were compared.At 24 hours,dry-wet weight ratio and the content of MDA in group D were lower than those in group C(P < 0 05),and the activities of SOD and GSH were increased(P < 0 05),and the degree of lung injury was alleviated.Compared with the indicators and lung tissue structure for the rest of the time period,there was no significant change between group D and group C.Conclusion: Bloodletting pretreatment of “Twelve well points of hand” can alleviate the lung tissue injury caused by oxidative stress reaction in a certain period of time,which mainly by mean of increasing the content of SOD and GSH in lung tissue,decreasing the content of MDA and the water content of lung tissue.
