| Objection:The reduction of male fertility is influenced by multiple factors,of which obesity is one of the causes.In this study,we established a mouse model of obesity reproductive disfunction,aiming to explore whether Huaji Jianpi Decoction can improve the quality of semen in obese mice through lipid-lowering,which will provides theoretical support for the clinical treatments of obese men with low fertility.Methods:1.One hundred mice of 3 weeks old were selected and adapted for one week under certain circumstances.Then they were randomly divided into two groups: normal group(30mice)and high fat group(70 mice).Normal group were fed with normal diet and high fat group were fed with high fat diet.After 10 weeks of feeding,the grease volume and blood lipid levels of the two groups were measured by Micro-CT and automatic biochemical analyzer to evaluate whether the obesity model was successful.In addition,the semen parameters of mice were detected by a sperm mass analyzer,the organ index was calculated after calibration with an electronic balance,and the ultra-structure of the tests were observed by transmission electron microscopy.2.10 normal mice were Selected as control group,fifty obese mice were randomly divided into model blank group,orlistat group,low dose group,middle dose group and high dose group(10 mice in each group).Distilled water was administered to both the control group and the sham group,and the drug group was administered with the corresponding concentration of the drug.After 6 week,the fat volume of the mice was measured by the Micro-CT.The serum lipids of mice were detected by automatic analyzer.The mRNA expression levels of PPAR-γ and C/EBP-α in liver tissues of mice were detected by RT-qPCR.3.Prepare sperm suspension and testicular sections,the quality of semen was detected by sperm mass spectrometer.The ultrastructure of testes was observed by transmission electron microscopy.The mRNA expression levels of TNF-α and MCP-1 in testis weredetected by RT-qPCRResults:1.After 10 weeks of feeding,the body weight,fat volume,and blood lipid levels(TC,TG,HDL,LDL)of the high-fat group were higher than those of the normal group,which proved that the obese model was successful;Moreover,by measuring the testicular wet weight,sperm density,sperm motility and other parameters,it was found that the semen quality and testicular wet weight of obese mice were lower than normal mice;Further observation of the ultra-structure of mouse testis revealed that the basement membrane of the testis of obese mice was accompanied by vacuolization,and a large number of vacuoles were observed in the cytoplasm.2.Corresponding gavage treatment was performed on the normal group,the sham group,and the drug administration group;The results showed that compared with the normal group,the sham group had high body weight,fat volume,blood lipid level and PPAR-γ and C/EBP-αmRNA expression levels;Compared with the model group,the indicators of the drug-administered group decreased to varying degrees.3.Sperm quality analyzer showed that the semen quality and testicular wet weight of obese mice were lower than those of normal mice;The ultra-structure of testicular tissue was observed by transmission electron microscopy,it was found that the number of mitochondria in spermatogenic cells was decreased and the apoptosis of spermatogenic cells was observed in obese mice;RT-qPCR was used to detect the expression levels of TNF-α and MCP-1mRNA in mouse testis.It was found that the expression of inflammatory factor mRNA in obese mice was higher than that in normal mice.Conclusions:1.Obesity can reduce the semen quality of mice,trigger germ cell apoptosis,and lead to decreased fertility in mice.2.Both orlistat and Huaji Jianpi Decoction could effectively decrease blood lipid level,inhibit the production of PPAR-γ and C/EBP-α adipokines.Moreover,those controlled the rapid growth of body weight,and the effect was most optimal in high dose group.3.Obesity could up-regulate the expression of TNF-α and MCP-1 mRNA in mouse testis,which could cause an inflammatory response and lead to the apoptosis of spermatogenic cells,reducing semen quality.The drug administration group can improve the low quality of semen induced by obesity to different degrees,and the effect was most significant in the high dose group. |
