Purpose: The purpose of the study was evaluated that Salidroside affected cell proliferation and further invested potential molecular mechanisms,as well as the sensibilization of the chemotherapy drugs Oxaliplatin and 5-Fluorouracil.Methods: Human gastric cancer cells SGC-7901 and MKN-45 were treated with different concentrations(10 ?M,20 ?M,40 ?M,80 ?M)for 24 h,and then CCK-8 assay detected cell proliferation.The wound healing assays were used to obverse migration ability.We selected the optimal concentration of salidroside is 80 ?M.TUNEL staining detected the apoptosis of gastric cancer cells.Extracellular acidification rate experiments can directly reflect the glycolysis rate of cancer cells,which can be used to evaluate the effect of salidroside on the metabolic capacity of cancer cells.The effects of salidroside on the expression of key rate-limiting enzymes in the glycolysis pathway of cancer cells were observed by qPCR,immunofluorescence and Western blotting.The cells were undertaken co-treatment using 80 ?M Salidroside and 20 ?g/mL Oxaliplatin,80 ?M Salidroside and 75 ?g/mL 5-Fluorouracil for 24 h.CCK-8 assay analyzed the sensitizing effect of salidroside conbine with two chemotherapeutic drugs.C57BL/6 mice were used to establish a subcutaneous xenograft model of gastric cancer,and different therapeutic ways(Using alone 12.5 mg/kg Salidroside,2.0 mg/kg Oxaliplatin,20 mg/kg 5-Fluorouracil,salidroside and Oxaliplatin,salidroside and 5-Fluorouracil)investigated the inhibiting effect of salidroside on gastric cancer cells.Results: The overview results suggest that different concentrations of salidroside inhibited the proliferation and migration in SGC-7901 and MKN-45 gastric cancer cells in a concentration-dependent manner and the best effect is 80 ?M(P<0.05).In addition,salidroside is responsible for nuclear fragmentation and apoptosis of gastric cancer cells.At the same time,salidroside inhibits the activity of human gastric cancer cells by inhibiting the expression of key proteins of glycolysis pathway in vitro.In vivo experiments showed that salidroside intervention in subcutaneous transplantation of tumors was significantly smaller than the control group.Salidroside combined with Oxaliplatin or 5-Fluorouracil can significantly decrease the gastric cancer cell viability.The volume of subcutaneous transplanted tumor mass has been decreased,showing a certain sensitizing effect.Conclusion: Salidroside inhibited the proliferation of SGC-7901 and MKN-45 gastric cancer cells by inhibiting the ability of glycolysis and reducing the migration ability of cells,induceing apoptosis,as well as improving the sensitivity of chemotherapeutic drugs.
