Objective: Colorectal cancer is one of the commonest malignant tumor.The annual incidence and mortality is ranked in the front line of malignant tumor.At present,the number of incidence and deaths between men and women are 1.21 and 1.17 in the world.The number of the incidence in developed countries are more than the developing countries.The incidence and mortality of colorectal cancer is increasing year by year.Taking into account the severity of colorectal cancer,new screening and evaluating methods appear to be in urgent need.The exist studies have shown that long non-coding RNA can affect the occurrence and development of tumor by participating in chromatin modification,transcriptional interference and intra-nuclear,or other related pathways,affecting the expression of genes in all aspects.These findings confirm the critical role of LncRNA in tumorigenesis of colorectal cancer.This study was designed to provide a new method for the diagnosis of colorectal cancer.Methods: We Collect 78 plasma of colorectal cancer patients before surgery and 76 plasma of the normal control.We detect the expression of 13 LncRNAs(PVT-1,UCA1,H19,RMRP,MALAT-1,CCAT1,CCAT1-L,CCAT2,HOTAIR,MALAT1,91 H,GAS5,MEG3)in 40 samples(20 plasma of colorectal cancer and 20 normal subjects)in Real-time RT-PCR test,and find the LncRNA express different in both groups.The LncRNA with different expression is screened out in the remaining samples.The relationship between the expression of LncRNA and the occurrence of colorectal cancer is investigated by comparing the expression of LncRNA in plasma.Thereby we obtain the LncRNAs with certain diagnostic significance.Results: The expression of LncRNA 91 H in plasma of colorectal cancer is higher than that in normal control.With statistical differences(P = 0.0231 <0.05).With the ROC curve AUC= 0.870>0.7(sensitivity = 86.2,specificity = 80.4,criterion>0.0095),91 H can act as an potential independently diagnostic indicator.The expression of LncRNA PVT-1 in plasma of colorectal cancer is higher than that in normal control.With significant statistical differences(P = 0.001 <0.01).With the ROC curve AUC=0.786>0.7(sensitivity = 69.0,specificity = 82.1,criterion>0.0165),PVT-1 can act as an potential independently diagnostic indicator.The expression of LncRNA MEG3 in plasma of colorectal cancer is higher than that in normal control.With significant statistical differences(P = 0.0002 <0.01).With the ROC curve AUC=0.819>0.7(sensitivity = 75.2,specificity = 82.1,criterion>0.163),MEG3 can act as an potential independently diagnostic indicator.As 91 H,PVT-1 and MEG3 are combined,the ROC curve AUC> 0.877(sensitivity = 82.8,specificity = 78.6,criterion>0.3797).So the three indicators can be used as a joint diagnostic criteria.Conclusion:LncRNA 91 H,PVT-1 and MEG3 in plasma of colorectal cancer patients express higher than that in normal human plasma.91 H,PVT-1 and MEG3 may act as independent diagnostic indicators for colorectal cancer.If 91 H,PVT-1 and MEG3 can be combined for detection,it may be more conducive to diagnose the colorectal cancer.
