Objectives:USP11 is an enzyme that regulates the stability of proteins through ubiquitination.VGLL4 is a transcription factor TEADs and inhibit the YAP gene,affect the Hippo pathway,thereby inhibiting the formation and growth of the tumor,the literature reported that VGLL4 protein in human tumor cells significantly down.However,it is unclear how the VGLL4 protein is regulated after transcription.The aim of this study was to investigate whether the ubiquitination of USP11 could regulate the growth,invasion and metastasis of renal cells by VGLL4.Methods: The subject of the first application of interaction through Western,Blot and immunoprecipitation verified USP11 and VGLL4 through the interaction of immunofluorescence and truncated design further validation of USP11 and VGLL4 and the combination of regional verification and to ubiquitination verification,and finally through the Si-USP11 for the renal carcinoma cell growth,invasion and metastasis effectResults: The interaction between USP11 protein and VGLL4 protein determines the binding domain of USP11 protein and VGLL protein.USP11 can stabilize the VGLL4 by ubiquitination,and promote the growth,invasion and metastasis of renal cell carcinoma after Si-USP11Conclusion:This study demonstrated that USP11 and VGLL4 interact with each other in renal cell carcinoma,and Si-USP11 may promote the growth,invasion and metastasis of renal cell carcinoma...
