Objective:In recent years,with the development of thrombolysis and catheterization technology,in order to avoid massive bleeding during cardiac and other vascular operations,blockade of blood flow at aorta abdominalis is often chosen at a specific stage,which results in temporary ischemia of spinal cord tissue,and secondary injury after blood supply recovery is called spinal cord ischemia-reperfusion injury.Sufentanil is widely used in clinical anesthesia as an opioid sedative and analgesic drug.A large number of literatures have confirmed that sufentanil has significant protective effect on tissue after ischemia-reperfusion,but few studies have been conducted on spinal cord.Therefore,the effect of sufentanil pretreatment on inflammatory factors MPO,IL-6 and IL-15 in spinal cord ischemia-reperfusion injury in rats was investigated,and the effect of sufentanil on inflammatory mechanism in spinal cord injury was discussed.Methods:Forty healthy SD male rats were randomly divided into sham operation group(Sham group,n=10),ischemia-reperfusion group(IR group n=10),sufentanil pretreatment 5?g/kg group(Suf group,n=10).Spinal cord ischemia-reperfusion model was prepared by occlusion of thoracic aorta with arterial clamp.The postoperative motor scores of rats were measured by Tarlov method,and the morphology of spinal cord histocyte was observed by HE staining.The expression of MPO in this kind of tissue was detected by Western Blot,and the contents of IL-6 and IL-15 in spinal cord histocyte of rats in each group were detected by ELISA.Results:Tarlov score in the IR group was higher than that of the sham group,and Tarlov score of the Suf group was lower than that of the IR group but higher than that of the sham group.Histological observation of the spinal cord: Under HE staining microscope,the spinal cord tissue of the sham operation group showed normal performance,displaying rounded and clear structure,coupled with complete and centered nucleus contour;Extensive denatured neurons,inflammatory cell infiltration,karyopyknosis deviation,fragmentation,and dissolution as well as vacuolization were spotted in spinal cord tissue of the ischemia-reperfusion group;The number of spinal cord tissue neuron damage necrosis in the Suf group decreased,with nuclear morphology being basically normal.Determination of MPO activity as well as content of IL-6 and IL-15 in spinal cord tissue: The contents of MPO,IL-6 and IL-15 in Suf group were all lower than those in the IR group,the difference of which was statistically significant(P<0.05);The contents of MPO,IL-6 and IL-15 in the IR group were all higher than those in the sham group,the difference of which was very obvious(P<0.05).Conclusion:Sufentanil can reduce the expression of MPO,IL-15,and 1L-6 in rats' tissues injured by spinal cord ischemia-reperfusion,reduce inflammation damage,and alleviate spinal cord ischemia-reperfusion injury.
