Objective: To investigate the therapeutic effects and the possible mechanism of regulatory signalling pathway of sulforaphane on 2,4-dinitrochlorobenzene(DNCB)-induced AD mouse model.Methods: Forty female Balb/c mice were divided into 5 groups(n=8 in each group):(1)negative control group: the mice were stimulated with blank buffer without DNCB,and an equivalent of PBS was intraperitoneally injected one hour later;(2)AD group:DNCB was given to induce atopic dermatitis and the same amount of PBS was intraperitoneally injected in one hour;(3)SFN2.5 group: DNCB was given to induce atopic dermatitis and the intraperitoneal injection of sulforaphane was given to the mice(2.5 mg / kg);(4)SFN5 group: DNCB was given to induce atopic dermatitis and the intraperitoneal injection of sulforaphane was given to the mice(5mg / kg);(5)SFN10group: DNCB was given to induce atopic dermatitis and the intraperitoneal injection of sulforaphane was given to the mice(10 mg / kg).The experiment period was 24 days,and the changes of skin lesions,pruritus scores,mouse ear thickness and dermatitis scores were measured.At the end of the experiment,the blood samples and the back lesions of the mice were collected.The Ig E levels of serum were detected by ELISA.The HE staining and toluidine blue staining were used to test the thickness changes,eosinophils and mast cell infiltration of skin tissues of the mice and Western Blot was used to detect the expression levels of NRF2 and HO-1 in the dorsal lesions.Results: 1.The weight of each group was normal and this phenomenon ensured that the SFN had no influence for mice.2.Compared with the negative control group,the AD group had obvious symptoms of dermatitis,and the thickness of the ear(P <0.05),the dermatitis scores(P <0.05),the pruritus scores(P <0.05),the thickness of skin epidermis (P <0.05),the eosinophil and mast cell infiltration(P <0.05)increased significantly,and the serum total Ig E level was increased as well(P <0.05).The SFN treatment group alleviated dermatitis symptoms,and the thickness of the ear(P <0.05),the dermatitis scores(P <0.05),the pruritus scores(P <0.05),the thickness of skin epidermis(P <0.05),the eosinophil and mast cell infiltration(P <0.05)decreased significantly than that of AD group.In addition,the expression of inflammatory antioxidants Nrf2 and HO-1 were increased(P <0.05),and the therapeutic effect is more efficient with the increasement of SFN therapeutic doses.There were significant differences between the treatment groups and the AD group(P <0.05)and a dose-dependent effects in SFN2.5 group,SFN5 group,SFN10 group for AD treatment(P <0.05).Conclusion: SFN has a certain therapeutic effect on DNCB-induced atopic dermatitis,and its mechanism may be related to the activation of NRF2/HO-1 signaling pathway.
