| Objective: To investigate the protective effect of a new compound probiotics on trinitrobenzene sulfonic acid-induced IBD mice,and whether its mechanism is related to improving intestinal mucosal barrier permeability and affecting the expression of tight junction protein occludin gene,and to compare the efficacy with that of Saccharomyces brasiliensis.Methods:SD mice aged 4-5 weeks were randomly divided into four groups: normal control group,TNBS model,TNBS + SB treatment group and TNBS + compound probiotics treatment group.Normal control group: no treatment,normal feeding;TNBS model control group: 2 days after modeling,0.5 ml sterilized saline was given orally once a day for 1 week;TNBS + SB treatment group: 2 days after modeling,0.4 ml SB + 0.1 ml sterilized saline was given orally once a day for 1 week;TNBS + compound probiotics group;2 days after modeling,0.4 ml compound probiotics + 0.1 ml compound probiotics were given orally.The normal saline sterilized with ml was given orally once a day for one week.The general state of mice,survival rate,body weight,stool status,gross condition of colon tissue were observed with naked eyes,and pathological changes of colon tissue were observed with HE staining.The expression of tight junction related protein 0cclludin-1 gene was detected by Real time PCR.Result:The blood stool and diarrhea symptoms appeared in the TNBS group,and the body weight gradually decreased in the first three days after the model establishment.The body weight increased with the relief of IBD mice’clinical symptoms,but the growth rate was smaller than that of the Brassica yeast group,the compound probiotics group and the blank control group.Disease activity index increased first and then decreased.After HE staining,there were obvious inflammatory cell infiltration and inflammatory mass in TNBS group,and a large number of destruction of intestinal epithelium and the structure of the pelvic fossa.In the normal control group,intestinal tissue developed well,intestinal epithelium was intact,and the structure of the crypt was normal.In TNBS model group,the intestinal epithelium and crypt structure were destroyed,and a large number of inflammatory cells infiltrated.After SB and probiotics were given,inflammatory cells infiltrated in colonic tissue,which was significantly less than that inTNBS group.RT-PCR results showed that the expression of tight junction-related protein occludin in TNBS+SB group and TNBS+probiotics group increased,but there was no significant difference compared with TNBS group.Conclusion: 1."Compound probiotics" has protective effect on intestinal tract of IBD mice.Its mechanism may be related to reducing intestinal permeability,promoting the expression of tight junction-related proteins and exerting the protective effect of intestinal mucosal barrier.2.The protective effect of probiotics on intestinal mucosal barrier was similar to that of yeast group. |
