Objective: As one of the most common malignant tumors in the world,lung cancer ranks first among all kinds of malignant tumors.The results published in the journal CA in November 2018 show that lung cancer is still the most common malignant tumor in the world,in which lung cancer occurs.11.6% of tumors are also the leading cause of cancer deaths,accounting for 18.4% of total cancer deaths.These data indicate that lung cancer is characterized by high morbidity and high mortality.Among them,non-small cell lung cancer(NSCLC)accounts for 75-85% of lung cancer.At present,lung cancer is mainly treated with surgery and supplemented by radiotherapy and chemotherapy.Although important results have been achieved in oncology research,recurrence and metastasis are still the main causes of treatment failure and death,and NSCLC is 5 years old.Survival rate does not exceed 15%,so finding effective therapeutic targets has become one of the breakthroughs in the treatment of lung cancer.Guanosine 5'-triphosphate-binding proteins(GTPBPs)are a class of proteins with signal transduction and have GTP hydrolase activity.GTP-binding proteins play an important role in cell signaling,cytoskeleton regulation,and protein synthesis.GTPBP2 is a member of the G protein superfamily,which interacts with components of the signal transduction pathway and may be involved in tumor development.At present,the relationship between the expression of GTPBP2 gene and tumor has not been reported in the literature at home and abroad.Methods: This experiment used Western blot to screen cell lines with high expression of GTPBP2 protein.The lentiviral vector-mediated RNAi technology was used to silence the GTPBP2 gene and the silencing effect was detected by Western blot and real time PCR.Cell scratch assay,transwell assay and CCK8 assay for changes in cell migration and proliferation after GTPBP2 gene silencing.Western blot was used to detect the expression of cell migration,invasion and proliferation related proteins after GTPBP2 gene silencing,and Wnt/?-catenin signaling pathways relevant protenins and the biological function of GTPBP2 in non-small cell lung cancer cell lines was further verified.Results: The expression of GTPBP2 protein in A549 cell line was higher than that in other cell lines.The migration ability of A549 cells after silencing GTPBP2 gene was lower than that of the negative control group.The invasive ability of A549 cells was significantly decreased after silencing GTPBP2 gene.Silencing GTPBP2 The proliferative capacity of A549 cells after the gene was lower than that of the negative control group.Conclusion: GTPBP2 is highly expressed in A549 and is associated with migration,invasion and proliferation of tumor cells.GTPBP2 may be involved in the occurrence and development of lung non-small cell lung cancer.
