Exosome is a kind of vesicle with a uniform size that cell active secrete,it has a lipid bilayer membrane structure.The diameter is about 40-100 nm and the density is about 1.10~1.18 g/ml.It was first found in the serum of sheep erythrocytes cultured in vitro.The exosomes secreted by different cells have different components and functions,and the main content is protein and RNA.Exosomes can be used as biomarkers for disease diagnosis.Exosome is naturally present in body fluids,including blood,saliva,urine and breast milk.Early studies suggested that exosomes perform protein transport functions,exchanging proteins and lipids.Until 2007,the researchers found that exosomes transport nucleic acids and participate in intercellular communication.Its proteins,RNA and fat are specific and carry important signaling molecules,expected to play a role in early diagnosis of a variety of diseases,it makes the space of exosome development rapidly expanding and becoming a hot research object.Astrocyte are glial cells in the brain that are about 3-5?m in diameter.Astrocytes are an important component of the nervous system,and they are interdependent with nerve cells during the development of the brain.By continuously cultivating astrocyte cells and collecting culture media,we can get the experimental materials to get exosome.By ultracentrifugation to secrete body outside,after detection by the method of electron microscope scanning secrete body outside the diameter of the form,scanning electron microscope(SEM)with energy to 1 to 30 kv electron beam,the printing on the surface of the sample to be analysed,and become by the electrons and the interaction between the surface electronic images,detect the exosome about 40 nm to 200 nm in diameter,finally through BCA method to detect exosome surface protein concentration,concentration suitable exosome in molecular screening for biological research level.In the rat stroke model,choose line bolt the rule of law as a rat model of cerebral ischemia reperfusion after intraperitoneal injection exosome,by observing the behavior of rats,and Zea Longa 4-point scale lack of standards for neurological signs of 5 rating,comprehensive evaluation of rat rat after recovery,analysis the secrete body in the role of stroke model in ratsThis study used the brain astrocytes of adult mice to extract and detect the exosome,by injecting intraluminal cerebral ischemia and reperfusion(MACO)model,observing the effect of exosomes on nerve damage animals,proving that the exosomes secreted by glial cells can help the functional recovery of neuro-damaged animals,explore the neurofunction recovery of exosomes in animals.It lays a theoretical foundation for the application of exosomes in clinical medicine.
