Objective:To investigate the effects of electroacupuncture at ST36 and DU1 acupoints on JNK-CREB-related signaling pathways in cerebral cortex of telomerase knockout(Terc-/-)mice and wild-type mice and their neuroprotective effects mechanism.Methods:Wild type mice were divided into blank group,ST36 group and DU1 group by random number table method.Terc-/-mice were divided into blank group,ST36 group and DU1 group with 6 mice in each group.All groups of mice were intervened once a day at a fixed time for 7 days.Wild type blank group and homozygous type blank group only simulated grasping movement;wild type ST36 group and homozygous type ST36 group electroacupuncture stimulated bilateral ST36 points,needling depth was about 1.5 mm;wild type DU1 group and homozygous type DU1 group electroacupuncture stimulated DU1 points,needling depth was about 6 mm.All the groups stimulated by electro-acupuncture were connected with bipolar connected needle by Han's electro-acupuncture instrument.The parameters were 2 Hz,2 mA,continuous wave,and the intervening time was 30 minutes.Morris water maze was used to detect the behavior of mice before and after intervention.The expression of JNK,CREB and p-CREB protein in cerebral cortex was analyzed by immunohistochemistry and Western blotting.Statistical methods of experimental data were compared between groups using one-way ANOVA.Result:?1?Behavioral test results:After intervention,the number of platform crossing in DU1 group was significantly higher than that in blank group?P<0.05?;In homozygous type,the number of platform crossing in DU1 group was significantly higher than that in blank group,the difference was significant?P<0.01?;In homozygous type,the number of platform crossing in DU1 group was significantly higher than that in ST36 group,the difference was significant?P<0.01?;In homozygous type,the number of platform crossing in DU1 group was significantly higher than that in ST36 group,the difference was significant?P<0.05?.In homozygous type,compared with blank group,DU1 group and ST36 group had a significant increase in the proportion of time in the central region,the difference was significant?P<0.05?.?2?The results of immunohistochemistry showed that in homozygous type,the expression of JNK protein in DU1 group was lower than that in blank group and ST36 group,the difference was significant?P<0.01?;the expression of CREB protein in DU1 group was higher than that in blank group,the difference was significant?P<0.001?;The expression of CREB protein in ST36 group was higher than that in blank group,the difference was significant?P<0.001?.The expression of p-CREB protein in ST36 group was higher than that in blank group,the difference was significant?P<0.01?;The expression of p-CREB protein in DU1 group was higher than that in blank group,the difference was significant?P<0.001?.?3?Western blot results showed that in homozygous type,the expression of JNK protein in ST36group was lower than that in blank group,the difference was significant?P<0.05?;The expression of JNK protein in DU1 group was lower than that in blank group,the difference was significant?P<0.01?;The expression of CREB and p-CREB protein in ST36 group was higher than that in blank group,the difference was significant?P<0.01?;The expression of CREB and p-CREB protein in DU1 group was higher than that in blank group,the difference was significant?P<0.01?;The expression of CREB and p-CREB protein in DU1 group was higher than that in blank group,the difference was significant?P<0.01?.The expression of CREB protein in DU1 group was significantly higher than that in ST36 group,the difference was significant?P<0.05?.In wild type,the expression of JNK and p-CREB protein in DU1group was lower than that in blank group,the difference was significant?P<0.01?;The expression of p-CREB protein in ST36 group was higher than that in blank group,the difference was significant?P<0.05?.Conclusion:?1?Electro-acupuncture ST36 and DU1 can improve the learning and memory ability of mice with telomere knockout.?2?Electro-acupuncture ST36 and DU1 inhibited the expression of JNK protein in the cerebral cortex of mice with telomer e knockout,but not in wild type mice.?3?Electro-acupuncture ST36 and DU1 could a ctivate the expression of CREB and p-CREB in the cerebral cortex of mice with telo mere knockout,but not in wild mice.
