Objective: In this study,the role of DNMT3 A in the development of gastric cancer was investigated by detecting the expression of DNMT3 A in gastric cancer,chronic atrophic gastric,juxtacancerous tissues and chronic non-atophif gastritis in Han and Hui nationalities in qinghai province,and its relationship between characteristics and clinical pathology of gastric cancer patients were analyzed.It is expected to provide theoretical and experimental basis for epigenetic mechanism and early diagnosis of gastric cancer.Methods: Gastric biopsy of 60 patients with gastric cancer and its juxtacancerous tissues,60 patients with chronic gastric atrophic and 60 patients with chronic nonatophif gastritis diagnosed by gastroscopy and gastric mucosal biopsy were collected.Among them,30 cases were Han and Hui in each group.At the same time,the case registration form was designed,and the information of each subject was collected in detail,including general data such as gender and age,and pathological data such as tumor size,depth of invasion,degree of differentiation and TNM staging.Tissues were analyzed by immunohistochemistry(IHC)method for the expression of DNMT3 A in each group.Results:1.The expression was statistically different(P<0.05)expression of DNMT3 A in the GC group was significantly different from that in the juxtacancerous tissue,CAG group and CNAG group(P<0.05),but there was no significant difference between the juxtacancerous tissues group,the CAG group and the CNAG group(P>0.05).2.The positive expression rates of DNMT3 A was statistically different in Han GC,paracancerous,CAG and CNAG(P <0.05).3.The positive expression rates of DNMT3 A was statistically different in Hui GC,paracancerous,CAG and CNAG(P <0.05).4.The positive expression rate of DNMT3 A in Han nationality GC,CAG CNAG,juxtacancerous tissues groupgastric mucosal biopsies were equal with that of Hui nationality at the same grade lesion(P>0.05).5.The protein expression level of DNMT3 A in GC group was correlated with the degree of tumor differentiation and lymph node metastasis(P<0.05),but there was no statistically differences among patient's gender,age,tumor size,depth of invasion,distant metastasis and TNM stage(P>0.05).Conclusions:1.The expression of DNMT3 A in GC tissues was sifnificantly higher than that in juxtacancerous tissues group,CAG and CNAG gastric mucosa tissues,it suggested that the up-relation of DNMT3 A expression may be related to the occurrence and development of GC.2.The expression of DNMT3 A in poorly differentiated GC tissue was sifnificantly higher than that in moderately and highly differentiated GC.The expression of DNMT3 A in GC tissues with lymph node metastasis was higher than that without lymph node metastasis.These results indicated that the high expression of DNMT3 A in gastric mucosa tissues may be involved in GC tumor differentiation,metastasis and development of GC cells.The dynamic detection of the expression of DNMT3 A in gastric mucosa tissues may provide certain reference to detecte the early GC,observe the therapeutic effect,and even judge the prognosis.3.There was no significant difference in the expression of DNMT3 A between the same organizations of Han and Hui nationalities,it may be suggest that DNMT3 A may not be an important factor causing the difference in detection rate between Han and Hui.
