Objective To study the protective effect of salidroside on myocardium in septic rats and explore its mechanism..Methods 96 clean-grade SD rats,male,were randomly divided into sham operation group(Sham group),sepsis group(CLP group),salidroside 30 group(SDS 30 group)and salidroside 60 group(SDS group 60),24 rats in each group,each took 6 to observe the7-day survival rate,and the remaining 18 rats were randomly divided into three subgroups of 6h,12 h and 24 h,with 6 in each subgroup.In the Sham group,the cecum was turned only after laparotomy,and the sepsis model was established by cecal ligation and puncture(CLP)in the CLP group,SDS 30 group and SDS 60 group.SDS 30 group received intraperitoneal injection of salidroside solution at a dose of 30 mg/kg within 30 minutes after operation;SDS 60 group received intraperitoneal injection of salidroside solution at a dose of 60 mg/kg within 30 minutes after surgery;Sham and CLP group Equal volume of normal saline was injected intraperitoneally.The rats in each subgroup were sacrificed at6 h,12h and 24 h after model establishment.Blood and tissue samples were collected.Serum IL-6,TNF-?,TLR-4,cTnI and BNP levels were detected by ELISA.Western Blot method The expression levels of TLR-4 and NF-?B p-p65 protein in myocardial tissue were detected.HE staining was performed on myocardial tissue,and its pathological changes were observed under light microscope.Results(1)Compared with Sham group,serum IL-6 and TNF-? levels were significantly increased in CLP,SDS 30 and SDS 60 groups at each time point(P<0.05).Compared with CLP group,SDS 30 at each time point.The levels of IL-6 and TNF-? in SDS 60 group were significantly lower than those in SDS group 30,and the levels of IL-6and TNF-? in SDS group 60 were significantly lower than those in SDS 30 group(P <0.05).(2)Compared with the Sham group,serum TLR-4 levels were significantly increased in the CLP,SDS 30 and SDS 60 groups at each time postoperatively(P<0.05),and TLR-4levels in the SDS 30 and SDS 60 groups at 12 h and 24 h.Compared with the CLP group,the point was significantly lower(P <0.05).At the 24 h time point,the TLR-4 level in theSDS 60 group was significantly lower than that in the SDS 30 group(P <0.05).(3)Compared with the Sham group,the levels of serum cTnI in the CLP,SDS 30 and SDS 60 groups were significantly increased at each time point(P<0.05).Compared with the CLP group,the levels of cTnI in the SDS 30 group were the same at each time point.The time point was significantly lower(P <0.05),and the level of cTnI in SDS group 60 was significantly lower than that in SDS group 30 at 6h and 12h(P <0.05).(4)Compared with the Sham group,the levels of serum BNP in the CLP,SDS 30 and SDS 60 groups were significantly increased at each time point(P<0.05).Compared with the CLP group,the BNP level in the SDS 30 group was significantly lower at 24 h(P<0.05),BNP level in SDS group 60 was significantly lower than that in SDS 30 group at24h(P <0.05).(5)Western Blot results showed that compared with Sham group,the expression of NF-?B p-p65 and TLR-4 protein in CLP group,SDS 30 group and SDS 60 group was significantly increased(P<0.05),compared with CLP group.The expressions of NF-?B p-p65 and TLR-4 in SDS 30 and SDS 60 groups were significantly decreased(P<0.05).(6)Histopathological changes of myocardial tissue: The pathological damage of Sham group was the least,and the pathological damage of CLP group was the heaviest.The pathological damage of SDS 30 and SDS 60 was significantly worse than that of Sham group,which was significantly reduced compared with CLP group.(7)Salidroside can improve the 7-day survival rate of septic rats.Conclusion During the development of sepsis,inflammatory factors are activated in large amounts,leading to myocardial damage.The protective effect of salidroside on myocardial injury may be through the inhibition of TLR-4/NF-?B/TNF-? inflammatory signaling pathway by salidroside,and the protection of the heart by down-regulating the level of inflammatory factors.
