Objective:To detect the protein expression of EETs synthase and hydrolysis enzyme and EETs amount in the mesenteric arterioles of rats in all groups.To verify that t-TUCB could decrease the portal pressure of PHT rats and to explore the underlying mechanism accounting for the effect of t-TUCB on reducing portal pressure by partly reversing myogenic response of mesenteric arteries from PHT rats.Methods:The model of portal hypertension was induced by subcutaneous injection of CCl4-olive oil.The portal pressure and blood pressure were measured by ALC-MPA multi-channel bio-signal analysis system.?2?The expression of EETs was measured by LC/MS/MS.The expression of CYP2C11 and sEH proteins in mesenteric tissues were detected by western blotting and immunohistochemistry,respectively.?3?The myogenic response and myogenic index of mesenteric arteries were measured by intraluminal mesenteric vascular perfusion.?4?The expression of eNOS protein in mesenteric microvessels was detected by immunoblotting.The myogenic response and myogenic index of mesenteric arteries were measured again after being incubated with L-NAME?a NOS inhibitor?.Results:?1?CCl4-olive oil method successfully represented liver cirrhosis PHT model,and t-TUCB can reduce portal pressure of PHT rats;?2?The expression of EETs/DHETs in the mesenteric arterioles of PHT rats was higher than that of the control group,while the t-TUCB could further increase the EETs/DHETs;?3?The myogenic response of the mesenteric arterioles in the liver cirrhosis rats was impaired,and t-TUCB was able to partially reverse the change of the myogenic response.t-TUCB can partially enhance the myogenic index of mesenteric artery in PHT rats.?4?t-TUCB partially reversed the myogenic response and reflected the better myogenic index of mesenteric arteries by down-regulating NO synthase in PHT rats.Conclusion:t-TUCB reduced the portal vein pressure of PHT rats by protecting myogenic response of mesenteric arterioles of PHT rats.t-TUCB reversed the myogenic response of the mesenteric arterioles of PHT rats by down-regulating the NO synthase.And the balance of EET and NO can partly explain the pathogenesis of PHT hyperdynamic circulation.
