Objective:To investigate the effects of gastrodin on oxidative stress and bone tissue damage in rats with fluorosis and its mechanism.Methods:72 healthy male Wistar rats weighing(140±5)g were selected.After normal diet,the rats were randomly divided into six groups: control group(basal feed,distilled water),gastrodin control group(basal feed,distilled water,400 mg /kg gastrodin),fluorosis group(basal feed,150mg/L Na F water),low dose gastrodin group(basal feed,150mg/LNa F water,100mg/kg gastrodin),medium dose gastrodin group(basal feed,150 mg /LNa F water,200 mg/kg gastrodin)and high dose gastrodin group(basal feed,150 mg/L Na F water,400 mg/kg gastrodin).Rats were weighed once a month during the experiment,and different doses of gastrodin were administered intragastrically after one month of fluoride(T1),two months(T2),three months(T3)and four months(T4),four rats were randomly selected,weighed and sacrificed,blood,bone and teeth were collected,serum Ca,MDA content,GSH-Px,CAT,ALP,TRAP and bone SOD activity and bone fluoride,dental fluoride content were observed.At the time of T4,the alveolar bone and femur HE staining were used to observe the pathological changes of bone tissue.The expression of Bax,Bcl-2,Caspase 3,9 and Cyclin D1 protein were observed by immunohistochemical staining of alveolar bone.Results:The serum SOD activity and GSH-Px activity in the fluorosis group were lower than those in the control group,the SOD activity in the bone tissue of the high-dose gastrodin group was higher than that in the fluorosis group,all were statistically significant(P <0.05).The levels of serum MDA in the fluorosis group were significantly higher than those in the control group(P <0.05).The serum CAT activity of the rats in the fluorosis group was lower than that of the control group.The serum CAT activity of the rats in the different doses of gastrodin groups were higher than that of the fluorosis group at T3 and T4,all were statistically significant(P <0.05).In the fluorosis group,the fluoride concentrations of bone and fluoride in the low,medium and high dose gastrodin groups were higher than those in the control group,some were statistically significant(P< 0.05);the fluoride concentration of bone and fluoride in the fluorosis group was significantly higher than that in each group.(P <0.05);compared with the gastrodin group,bone fluoride decreased gradually with increasing dose of gastrodin(P <0.05);there was a significant difference in fluoride concentration in high dose gastrodin group compared with medium and low dose gastrodin groups(P < 0.05).Compared with the control group,the thickness of the trabecular bone in the fluorosis group was increased,the thickness were unevenly distributed,the cell morphology was malformed,and the distribution was uneven.Compared with the fluorosis group,the cell morphology was restored,the thickness of the trabecular bone was restored,and it was strip-like,and the orientation was basically restored.Compared with the control group,the expression of Bcl-2 protein in fluorosis group decreased significantly,and the expression of Bax,Caspase 3,9 and Cyclin D1 protein increased significantly.However,the expression of Bcl-2 protein in gastrodin group increased significantly,and the expression of Bax,Caspase 3,9 and Cyclin D1 protein decreased significantly.Gastrodin,especially high dose(400mg/ kg),can reduce bone tissue damage in fluorosis rats and reverse up-regulation of Bax,Caspase 3,9 and Cyclin D1 protein expression and down-regulation of Bcl-2 protein expression in bone tissue caused by fluorosis.Conclusion :(1)Gastrodin can effectively alleviate the dental fluorosis and skeletal fluorosis caused by fluoride.(2)Excessive fluoride can decrease the antioxidant capacity of rats and change the levels of serum calcium,TRAP and ALP in rats,gastrodin can play a certain protective role.(3)Gastrodin regulates the expression of apoptosis-related proteins Bcl-2,Bax,Caspase3,9 and Cyclin D1 in bone tissue,and has a significant protective effect on bone tissue apoptosis,inferring that gastrodin may be a safe and effective treatment drug for fluorosis in rats.
