Objects: To explore the expressions of MicroRNA-652 in Esophageal squamous cell carcinoma(ESCC),and the correlation with the biological behavior of ESCC,such as proliferation and invasion,meanwhile,explore and verify its direct action sites.Methods: 37 pairs of esophageal squamous cell carcinoma tissues and adjacent non-tumor tissues were collected from patients with esophageal carcinoma who underwent surgical treatment.RT-qPCR was used to detect the expression of MicroRNA-652.In vitro experiments,RT-qPCR was used to detect the expression of MicroRNA-652 in human esophageal squamous cell carcinoma lines Kyse70,Kyse150,Te-1and ECA109,compared with normal human esophageal squamous cell line HET-1a.MicroRNA-652 mimics and MicroRNA-NC were synthesized and transfected into two cell lines with relatively low expression of MicroRNA-652,cell transfected for 48 h were collected,and the transfection efficiency was determined by RT-qPCR.CCK-8 assay was used to detect cell proliferation while Transwell invasion assay was used to detect cell invasion.On the database retrieval website,FGFR1 was found to be the most likely direct target gene of MicroRNA-652 in esophageal squamous cell carcinoma.The expressions of FGFR1-related mRNA or proteins were detected by RT-qPCR and Western-Blot in clinical samples.In vitro experiments,Luciferase reporter detected the connecting sites of FGFR1 and MicroRNA-652,Cells transfected for 48 h were collected,the expression of FGFR1's related mRNA and protein was detected by RT-qPCR and Western-Blot.Two cell lines were transfected with overexpressed FGFR1 plasmid,and the transfection efficiency was detected by Western-Blot to observe the changes of proliferation and invasion ability of MicroRNA-652-mediated cells.Results: The expression level of MicroRNA-652 in ESCC tissues is significantly lower than that in adjacent non-tumor tissues(P<0.05),FGFR1 related mRNA and protein levels are decreased(P < 0.05).In vitro,MicroRNA-652 expression level was significantly decreased in four ESCC cell lines compared with that of HET-1a(P<0.05),and the decrease was most obvious in two cell lines TE-1and Eca109.Compared with the blank control group,the proliferation and invasion ability of MicroRNA-652 mimics transfected group are decreased(P<0.05),and the levels of FGFR1-related mRNA and protein are decreased(P<0.05).Overexpression of FGFR1 can partially eliminate the inhibitory effect of MicroRNA-652 on the proliferation and invasion ability of ESCC cells(P<0.05).Conclusions:MicroRNA-652 is low-expressed in esophageal squamous cell carcinoma cells.MicroRNA-652 inhibits the proliferation and invasion of esophageal squamous cell carcinoma cells by targeting FGFR1.
